Invasion and metastasis are the main reasons for the death. Most ofthe patients with gastric cancer are in advanced stage when diagnosed andlose the optimal treatment opportunity. Thus to search the sensitive andspecial indexes that can reflect the biological behaviour of gastric cancerseems more important for precise clinical diagnosis.This article is intended to discuss the relationship between the proteinexpression of β-catenin, ki67and Her-2/Neu and the occurrence,development,biological behaviour and pathologic characteristics of gastriccancer; analyze the relationship between β-catenin and ki67and theprotein expression and gene amplification of Her-2/Neu to find reliableindexes for screening the gene state of Her-2/Neu by detecting theexpression of β-catenin,ki67and Her-2/Neu in the gastric cancer.In this experiment101cases of gastric cancer specimens werecollected as the experimental group while76cases of normal gastricmucous membranes as the control ones. β-catenin, ki67and Her-2/Neuwere detected respectively by immunohistochemistry SP method in thegastric cancer tissues and the proteins of different clinical pathologiccharacteristics.The gene state of Her-2/Neu in primary focus of gastric cancer wasdetected by fluorescence in situ hybridization (FISH).The data were analyzed by SPSS17.0;the variability was tested bychi-square test and Fisher exact probability and the correlation among theindexes was analyzed by Spearman correlation coefficient the differencehad statistical significance when P was less than0.05. The results showed as follows:1Detecting by Immunohistochemistry methodβ-catenin, ki67and Her-2/Neu were detected respectively byimmunohistochemistry S-P method in101cases of gastric cancer primaryfocus and the expression rates were74.3%,80.2%and15.8%respectively,which were much higher than those in the control groups.The differencebetween the experimental group and the control had statistical significance(p<0.05). The Spearman analysis indicated that the expression of β-catenin, ki67and Her-2/Neu had no significant difference in differentgender, different age and different tumor diameter of gastric cancer patients(p>0.05) while β-catenin and ki67had obvious significant difference indifferent differentiation level,different invasive depth and lymph nodepositive or negative group (p<0.05). β-catenin had no significantdifference in different TNM staging group (p>0.05), ki67had obvioussignificant difference in different TNM staging group (p<0.05) andHer-2/Neu had obvious significant difference in different differentiationlevel and TNM staging group (p<0.05) and had no significant differencein different invasive depth and lymph node positive or negative group (p>0.05).The correlation analysis indicated that the protein expression of ki67and Her-2/Neu had obvious correlation (r=0.567,P<0.05) while the proteinexpression of β-catenin and Her-2/Neu had no correlation and statisticalsignificance(r=0.478, P=0.116,P>0.05).2Detecting by fluorescence in situ hybridizationIn the primary gastric cancer focus, the gene amplification ofHer-2/Neu was not found in the85Immunohistochemistry stainingnegative or1+positive cases while there was Her-2/Neu amplification inthe16Immunohistochemistry staining2+or3+positive cases and the scoreascended with the conformity rate. The expression of ki67and theHer-2/Neu amplification had obvious significant difference (r=0.304, P<0.05) while the expression of β-catenin and the Her-2/Neuamplification had no significant difference (P>0.05).Conclusions by the experiment:1The expression of β-catenin, ki67and Her-2/Neu in the gastriccancer tissue is much higher than that in the control, which indicated thatthe indexes above-mentioned have close relationship with the occurrenceand development of gastric cancer.2The expression of β-catenin, ki67and Her-2/Neu have closerelationship with the clinical pathologic characteristics and the jointdetection of the indexes can provide full-scale reference for understandingthe tumor character, clinical diagnosis and prognosis judgment.3The expression of ki67and the Her-2/Neu amplification state hadobvious significant difference while the expression of β-catenin and theHer-2/Neu amplification state had no significant difference.
