| Objective: Breast cancer is one of the most common malignancies for women aroundthe world. Mentality and emotion play an important role on the incidence and developmentof tumor. Adrenergic neurotransmitter involved in tumor growth and metastasis. In thisstudy, we will test the expression of β2adrenergic receptor (β2-AR) protein in breastinvasive ductal carcinoma, so as to investigate the clinical significance of β2-AR in breastcancer.Moreover, to have further investigation on the biological behavior and reveal thepossible functioning mechanism of β2-AR in breast cancer through the discussion on thecorrelation expression of β2-AR, HER-2, and K-ras gene.Method: Checked the expression of β2adrenergic receptor, Her-2and K-ras geneprotein in10cases of normal breast tissue and33cases of invasive ductal carcinoma inparaffin by means of Immunohistochemical staining streptomycin avidin and Peroxidase(SP), to analyze and understand its clinicopathological significance for exploration thebiological behavior of breast cancer. Experimental data Processing and analysis were madeby application of SPSS17.0statistical software, nonparametric rank sum test(Mann-Whitney test), and spearman rank correlation test statistical methods.Results:1. expression of β2adrenergic receptor (β2-AR)β2-AR immunohistochemical positive reaction was at the cell membrane andcytoplasm, appeared as brownish yellow or brown granules. The β2-AR positiveexpression for10cases of normal breast was7/10; while the positive expression for33cases of breast cancer tissue is33/33. Differences in expression levels between the twoβ2-AR was significant (value Z=-4.876, value P=0.000, p <0.05).2. Her-2immunohistochemical positive reaction was at the cell membrane, appearedas brownish yellow or brown granules. The Her-2positive expression for10cases of normal breast was2/10; while the positive expression for33cases of breast cancer tissuewas33/33. Differences in expression levels between the two Her-2was significant (valueZ=-2.752, value P=0.006, p <0.05).3. K-ras immunohistochemical staining was at the cell membrane, appeared asbrownish yellow or brown granules. The K-ras positive expression for10cases of normalbreast tissue was3/10; while the positive expression for33cases of breast cancer tissuewas28/33. The difference between the two K-ras expression level was significant (valueZ=-3.126, value P=0.020, p <0.05).4. Study on correlation of expression of β2adrenergic receptor with Her-2and K-rasIn the study on correlation of expression level of β2-AR with Her-2and K-ras inbreast cancer tissue, the expression of β2-AR and Her-2showed positive correlation(value r=0.302, P=0.024, p <0.05) and the expression of β2-AR and K-ras showednegative correlation (value r=-0.286, P=0.025, p <0.05),which of statisticalsignificance.5. The relationship between the Expression of β2-AR and clinical and pathologicalfeaturesExperimental data showed that the expression of β2-AR differences between lymphnode metastasis in clinical pathology parameters were significant, and differences betweenage, tumor size, clinical stage, histological grade and hormone receptor in each group hasno statistical significance.Conclusion:(1)β2-adrenergic receptor (β2-AR), Her-2and K-ras expression exist in both normalbreast tissue and breast cancer tissue, and expression in breast cancer tissues wassignificantly higher than in normal breast tissue, indicating that β2-AR, Her-2, K-rasexpression are all associated with the occurrence and development of breast cancer.(2)The expression of β2-AR differences between lymph node metastasis in clinicalpathology parameters were significant, and differences between age, tumor size, clinicalstage, histological grade and hormone receptor in each group has no statistical significance.It indicated that potential risk of lymph node metastasis is also large for the breast cancerwith high β2-AR expression.(3) Expression of β2-AR and Her-2showed positive correlation; expression of β2-ARand K-ras was negatively correlated. It indicated that in the process of β2-AR in breast cancer may play a role through Her-2and K-ras. |
