Objective:Liver cancer is a malignant tumor that occurred in the liver, it is also one of the mostcommon malignancies in clinical. In the currently known malignant disease, the incidenceof liver cancer ranked sixth in the world. In high incidence areas like eastern Africa and thePacific coast, the incidencecan reach30/100000, while in Australia, America and otherincidence areas, can reach about5/100000. It ranked third in mortality of malignantdisease, up to20/100000, and in10years, the incidence and mortality rates showed aslow upward trend. In recent years, along with constantly updated surgical techniques,likeuniversal application of chemotherapy, targeted drug development and treatment ofimmune therapy and other kinds of new technologies, liver cancer mortality relative to theprevious has improved a lot, but long-term survival of patients with hepatocellularcarcinoma with tumor survival and recurrence of the situation is still dissatisfied. This isbecause the incidence of liver cancer in early clinical symptoms are not obvious, and thepatients only found liver cancer in the conditions of jaundice, fatigue, and other symptomsof ascites to do the relevant checks.Unfortunately, they often have lost the opportunity ofsurgical treatment, the proliferation of cancer cells already exist, the prognosis has beenunsatisfactory. So, early detection, early diagnosis, early treatment of liver cancer is amatter of general interest. Research on the long non-coding RNA open a new type of idea,there are many documented, long non-coding RNA plays an important role in a variety offunctions in the human body. In this article, we want discuss the role of the development ofa long non-coding RNA DHC occur in liver cancer, study its mechanism of action and theoccurrence of related functions, to evaluate its possibilities as a target for early diagnosis ofliver cancer.Methods:In previous experiments, we used tissue microarrays of patients from the EasternHepatobiliary Surgery Hospital of liver cancer with hepatocellular carcinoma tissuesamples and adjacent tissues samples to found a long-chain non-coding RNA-AK123488(NCBI number), we named it the long chain of non-coding RNA DHC, there is a clearcut expression in HCC. This raises the interest of our research on them. To determine ifthis phenomenon is not accidental, we examined the comparative another42pairs ofhepatocellular carcinoma in patients with paraneoplastic tissue differences in theexpression levels of LncRNA-DHC by PCR to verify this phenomenon, and we have done a normal the expression of liver tissue and HCC LncRNA-DHC contrast. Found thatthe expression of this long chain of non-coding RNA in HCC tissues than in normal livertissue was significantly reduced. This shows that the expression of this long chain of non-coding RNA in the occurrence and development of liver cancer does have a significantimpact, or, we guess, if it also plays an important role in the development of liver cancer.To further investigate the function and mechanism LncRNA-DHC, we used5’,3’-RACE method to obtain the full-length sequence LncRNA-DHC and PCR experimentsto conducted that there is a low expression to verify this long chain of non-coding RNA inliver cancer cells. Then we picked out85pairs of patients who had complete clinical andpathological data, through the expression of PCR into high expression group and the lowexpression group, and as a basis for LncRNA-DHC prognosis and patient survival analysisconducted.We LncRNA-DHC conducted a preliminary exploration of the function. Knockdown the expression LncRNA-DHC, and then transfected by lentivirus way inhepatocellular carcinoma cell line HCC LM3, SMMC-7721in interfering with theexpression of LncRNA-DHC. After the formation of stably transfected cell lines culturedusing a CCK8assay for hepatoma cell lines in vitro proliferation ability of LncRNA-DHCin front interference. In order to verify this effect, we then build a model of tumor-bearingnude mice, the stably transfected cell lines implanted subcutaneously into nude mice. Wefound that knock-down the cancerous tissue LncRNA-DHC cells formed significantlylarger than the control group. Then, we use the same cell lines discussed the impactLncRNA-DHC for metastatic liver cancer cells through Transwell experiments, we foundthat knock-down LncRNA-DHC in SMMC-7721and HCC LM3cell lines have beensignificantly enhanced metastasis.Results:(1)Expression levels of LncRNA-DHC in cancer tissue were significantly reduced.(2)LncRNA-DHC was related with nodule and microvascular invasion, alsorelated with the patient’s prognosis after surgery(3)In LncRNA-DHC high expression cell line HCC LM3, SMMC-7721before andafter the change in interference detection interference after CCK-8cellproliferation LncRNA-DHC found LncRNA-DHC suppress the proliferationand migration of HCC cells in vitro.(4)Transwell experiment found that knock-down LncRNA-DHC in HCCLM3and SMMC-7721cell lines have been significantly enhanced metastasisResultsExpression levels of LncRNA-DHC in cancer tissue were significantly reduced and itwas related with nodule and microvascular invasion, also related with the patient’sprognosis after surgery, knock-down LncRNA-DHC can affect proliferation and invasionof hepatoma cells.
