Objective: Morphology of neointima, expression of TGF-β1/Smad signaling proteins and MSCs markers inneointima were observed in common carotid artery balloon injured model of rats, to explore thecorrelation between them and their relationships with the neointimal formation. This study lay a goodfoundation for further studying mechanism of TGF-β1/Smad signaling pathway affecting the developmentof neointimal after vascular injury.Methods: Common carotid artery balloon injured model of rats were built.42male SD rats weighted350g-400g were randomly assigned into blank control group and6groups underwent left carotid arteryballoon injury(0h,1d,3d,7d,14d,21d). Arteries underwent intervention were harvested at0hour,1day,3days,7days,14days and21days and subjected to immunohistochemistry and western blotting analysisto observe protein expression of TGF-β1/Smad signaling pathway and MSCs markers. Calponin tested asthe smooth muscle marker, Ki-67as proliferation marker and Vimentin as mesenchyme were tested aswell.Results:(1)There were no significant neointima formation in control,0h,1d,3d groups, obvious neointimawere observed in7,14and21d groups(P<0.001).(2) TGF-β1/Smad signaling proteins were detected byimmunohistochemical staining and western blotting, and there were no significant staining in in control,0h,1d,3d groups, but expressed obviously in7,14and21d groups,protein expression reached the peakat14d groups.(3)Nestin and CD29were tested as MSCs markers by immunofluorescence, there werepositive cells covered on neointima in7and14d groups, and the positive cell rates are Nestin(0.01±0.01,0.1±0.02),CD29(0.01±0.01,0.09±0.01).(4) Protein expression of Calponin、Vimentin and Ki-67weredetected byimmunohistochemical staining, and there were no significant staining in in control,0h,1d,3dgroups,2but expressed obviously in7,14and21d groups with positive rates Calponin(0.36±0.02,0.88±0.05,0.15±0.01) Vimentin(0.28±0.01,0.85±0.07,0.1±0.01)and Ki-67(0.3±0.05,0.87±0.04,0.08±0.01).Conclusion:(1) Common carotid artery underwent balloon injury could formation neointima.(2)Higherprotein expression of TGF-β1/Smad signaling proteins and MSCs markers in neointima indicate the...
