Objective: To explore the action of TGF-β1/Smad signalpath in the occurrence of intestinal adhesion by determining the mRNAand protein expression of transforming growth factor–β1(TGF-β1),P-Smad3, P-Smad7, E-cadherin, Integrin Linked Kinase (ILK), α-smooth muscle actin (α-SMA).Methods: A total of90SD rats weredivided into Group A, Group B and Group C, and there were30rats ineach group. Rats in Group C were made the models of intestinaladhesion, rats in Group B only were received the surgery of opened andclosed the abdomen, and rats in Group C didn’t received any surgery. Atotal of10rats were killed on2nd,5thand8thday after surgery and thecaecum samples were removed. The protein level of TGF-β1, Smad3,Smad7and E-cadherin in each group were determined by SP methods;the protein level of ILK and α-SMA in each group were determined byWestern-blot; the mRNA level of TGF-β1, Smad3, Smad7, E-cadherin,ILK and α-SMA in each group were determined by RT-PCR.Results:(1)The adhesion tissue, proliferation of fibrous tissue and inflammatorycell of infiltration didn’t occur after surgery in the rats of Group A andGroup B. The adhesion of caecum to colon, small intestine andabdominal wall occurred surgery in the rats of Group C, proliferation offibrous tissue and lots of inflammatory cell of infiltration appeared inthe adhesion tissue. Glassy degeneration even could be seen in the fibertexture.(2) The mRNA and protein expression of TGF-β1, P-Smad3, P- Smad7, E-cadherin, ILK and α-SMA in the rats of Group A and GroupB on2nd,5thand8thday after surgery were not significantly different(P>0.05), and those in the rats of Group C on2nd,5thand8thday aftersurgery were significantly different (P<0.01).(3) The mRNA andprotein expression of TGF-β1, P-Smad3, P-Smad7, E-cadherin, ILKand α-SMA in the rats of Group A were not significantly differentcompared with those in the rats of Group B (P>0.05), but the mRNAand protein expression of TGF-β1, P-Smad3, P-Smad7, E-cadherin,ILK and α-SMA in the rats of Group C were significantly differentcompared with those in the rats of Group A and Group B(P<0.01).Conclusion:(1) The over expression of TGF-β1and ILK isprobably related to the intestinal adhesion;(2) The abnormal expressionof Smad3and Smad7is probably related to the intestinal adhesion;(3)The reduced expression of E-cadherin and increased expression of α-SMA is probably related to the intestinal adhesion;(4) TGF-β1/Smadsignal path probably takes part in the occurrence of the intestinaladhesion. |
