| Aim: To extract peptides according to in vitro anti-tumoractivity-orientation from chinese medicine Gecko (Gecko swinhoana Gunther.).The peptides structures of higher anti-tumor activity and purity wereelucidated through de novo method of MS/MS.Methods: Semi-bionic extraction performed on Gecko peptidesextract.Anti-tumor activity in vitro MTT assay after separation andpurification were detected to choose a higher anti-tumor and purity peptideto use of tandem mass spectrometry and NMR spectroscopy for structuralanalysis.Results: Using Sephadex G series and HPLC etc to purify eight peptides,in vitro screening showed that8peptides had different anti-tumor activityto breast (MCF-7), lung (A549), ovarian cancer (SKOV3), the peptides showedbetter activity than raw peptides. Their purity were checked by HPLC, peptideNO.8were choose to be elucidated structure de novo through MS/MS methodThe peptide structure may be speculated as follows: Conclusion:The research shows that gecko peptides have broad-spectrumanti-cancer activity and slight selectivity of certain cancer.A unknownoligo-peptide structure were elucidated through de novo MS/MS method underthe condition of gen-bank data absence of gecko. |
