Objective:This study is to isolate and identify an Entamoeba strain from macaques and to conduct a detailed examination of its bionomics.Methods:Stool samples of macaques which contains cysts of Entamoeba nuttalli were obtained from Guizhou Province. These samples were xenically cultured in Robinson’s medium, and named as GY4. The E. nuttalli strain GY4was successfully cloned and cultured axenically. Genomic DNA was extracted from the trophozites of axenic cultures. PCR amplifications of the serine-rich protein (SRP) and18S rRNA genes were performed and subjected to sequencing. The virulence of the Entamoeba nuttalli isolate was evaluated in vitro and in vivo.Resualts:Regions of18S rRNA gene were amplified by PCR and sequenced genes of E. nuttalli GY4isolates show some differences comparing with E. histolytica. These genes we selected is suitable for the classifition of Entamoeba spp. Analyses of virulence suggested that the E. nuttalli may have different pathogenic mechanism comparing with E. histolytica.Conclusion:E. nuttalli GY4strain showed several factors correlating to the virulence which was seen in E. histolytica. Virulence analysis suggested that the E. nuttalli GY4strain was different to E. histolytica. This may attribute to the low expression levels of pathogenic related genes of E. nuttalli GY4strain. Further study including genetic analysis should be necessary to assess its virulence.
