[Objective]To investigate the effect of Huxinkang on expression of matrix metalloprotei nase-9and matrix metalloproteinase tissue inhibitor-1(TIMP-1) in thoracic ao rta of rabbits with atherosclerosis.[Methods]Thirty six male rabbits were randomly divided into four groups:normal contr ol group (treated with normal diet),pathological control group,Huxinkang co ntrol group and simvastatin control group cholesterol diet for16weeks. Kill ed one rabbit randomly in the model group at the end of12th week, and ob served the sample under the light microscope and discovered the rabbit mod el of atherosclerosis Established Successfully, Then Huxinkang control group treated with Huxinkang(1.08g/kg·d) and simvastatin control group treated w ith simvastatin(2.5mg/kg·d) for four weeks. After16weeks, taking the ser urn of rabbits to test TC TG HDL-C LDL-C and staining aortae by HE to o bserve the morphologic change, The mRNA expressions of MMP-9and TIMP-1on aortae were examined by semiquantitative reverse transcription polymer ase chain reaction (RT-PCR).[Results]Compared with the control group, the thoracic aortae lipid plaque of the mo del group formed significantly, the other group formed hardly, and that show s the model atherosclerotic rabbits established successfully. The expression o f MMP-9in four-group were0.36±0.18、0.85±0.15、0.71±0.19、0.57±0.13, compared with model group, respectively, with significant difference amon g them(P<0.01). The expression of TIMP-1mRNA in above stated in four- group were0.38±0.15、0.72±0.13、0.85±0.14、0.93±0.19, the expression o f TIMP-1mRNA in Huxinkang control group and simvastatin control group were more than that of pathological control group(P<0.05, P<0.01).[Conclusion]Huxinkang can reduce MMP-9level,increase TIMP-1level, inhibit the format ion of atherosclerosisin in thoracic aorta of rabbits with atherosclerosis.
