Silk fibroin is a natural polymer fibrin, the ionic liquid is a new environmentallyfriendly green solvent, it became fibroin processing, and integrated applicationprovides new ways and ideas. A series of basic ionic liquids were synthesized and an[Bmim]AC ionic liquid with good dissolving capacity for silk fibroin was selected.The dissolving properties of silk fibroin in the [Bmim]AC ionic liquid were researched.The results showed that under75℃, the largest dissobulity could reach to15%. Thestructures of nature and regenerated silk fibroin were characterized by X-raydiffraction (XRD) and Fourier transform infrared spectroscopy (FT-IR), silk fibroin inthe dissolution process doesn’t derivatization. Therefore,[Bmim]AC ionic liquid is theexcellent solvent for silk fibroin. The dissolve mechanism of silk fibroin in the[Bmim]AC was analysized.The rheological properties of silk fibroin/ionic liquids were also studied. Theresults showed that: With the concentration increasing, flow type from Newtonian intono-Newtonian flow, the shear rate direction toward a low rate, the concentration is toohigh, the system mobility were decreased. The concentration was reached15%, theviscous flow activation energy37.56KJ/mol, the concentration was reached15%,viscous flow activation energy41.15KJ/mol. With the concentration increasing, theviscous flow activation energy was increased, the temperature was sensitived. The highconcentration solution was tested, the stability of the solution temperature was ensured.With temperature increasing, the apparent viscosity of the solution was decreased, thecritical shear rate direction toward the high shear rate. Within a certain frequency range,the solution gel point was not generated, the solution have a good performance andstability of. The concentration was reached15%, its have good for film wasproducted. The regenerated silk fibroin films from ionic liquid was prepared by using glassscraping and ethanol as coagulant. Under the optimal conditions, the regenerated silkfibroin film was of better properties than the traditional film and the properties were asfollows: the breaking strength12.72MPa, swelling rate23.25%, porosity45.05%. Theregenerated film was tested by SEM, FT-IR, XRD, TGA. The results showed thatfibroin in the ionic liquid is dissolved directly and without derivatization reactionsoccur, and regenerated film contains two structures SilkⅠ and SilkⅡ. However,compared with natural silk fibroin, the thermal stability of regenerated film declined.By MTT assay, The biocompatibility of regenerated film was tested. The resultsshowed that the number of regenerated films viable cells significantly more thantraditional film. In a certain range of concentrations, the regenerated film material canbe applied in the cell. Cells were able to adhesion by growing on the film and have agood biocompatibility. In the biological field, film materials were provided a realisticbasis for further research and application.
