Lingzhi, also known as Lingzhicao or Hongzhi or Chizhi, which belongs to Ganoderma genus, Polyporaceae. There are108species in China, including Ganoderma tsugae Murr, Ganoderma liPsiense(Batseh)GF.Atk, Ganoderma duropora, and so on. However, it is stated in the Chinese pharmacopoeia, which is the fruiting body of Ganoderma lucidum (Leyss.ex Fr.)Karst or Ganoderma sinense Zhao.Xu et Zhang. Ganoderma lucidum have been used as pharmaceutical fungi in folk for a long history. They are fungi widely distributed in China, Japan and Korean Peninsula. Ganoderma lucidum spore is the germ cell collecting from Ganoderma lucidum during the period of maturity. Modern medical and pharmacological researches have proved that the spore was active in improving immunity, resisting tumor, antivirus, eliminating inflammation, and so on. It is the adjuvant therapy medicine for the cancer patients in clinic. It can significantly alleviate the decline of white blood cell, reducing the side effect of chemotherapy, to extend the survival time of patients.In recent years, the researches on chemical compositions in Ganoderma lucidum spore have gained more and more attention. To add new content to the systematic studies of the chemical compositions in Ganoderma lucidum spore and facilitate its progress, as well as establish foundation for its further pharmacological studies and clinical application, this thesis made further investigation and exploration on the chemical compositions in Ganoderma lucidum spore on the basis of previous studies.The Ganoderma lucidum spore oil was extracted from the Ganoderma lucidum spore by the method of supercritical carbon dioxide extractive technology. With the used of modern chromatography techniques on silica gel, ODS, Sephadex LH-20and preparative HPLC, the oil extraction led to the isolation of8compounds. Among them,7compounds were identified by means of physical and chemical methods and spectral data analysis, which includes EI-MS、HR-ESI-MS、GC-MS、1H-NMR、13C-NMR、DEPT. These compounds are1,3-dipalmitoyl-2-oleoyl-glycerol (Ⅰ),1-stearoyl-2-oleoyl-3-palmitoyl-glycerol (Ⅱ),1,2-dioleoyl-3-palmitoyl-glycerol (Ⅲ),1,2,3-trioleoyl-glycerol (Ⅳ),methyl hexacosanoate (Ⅴ), ergosterol (Ⅵ), ergosterol peroxide (Ⅶ) respectively. Compounds Ⅰ~Ⅴ were first obtained from Ganoderma.We develop a quantitative method for determination of1,2,3-trioleoyl-glycerol in the Ganoderma lucidum spore oil by HPLC-ELSD. The method of HPLC-ELSD is simple and accurate, repeatable and can be used for determination of Ganoderma lucidum spore oil.In addition, recent advances in the research of chemical constituents and pharmacological roles of Ganoderma lucidum and the spore of Ganoderma lucidum were reviewed.
