| The incidence of facial malformations situates the first place of the neonatal congenital malformations. As polygenetic susceptibility to the disease, facial malformations are caused complexly. The first branchial arch(mandibular arch) including the lower jaw jaw, jaws and teeth, such as organ, mandibular growth is the result of develpoment by the interaction between epithelial and mesenchymal, and a variety of signaling molecules and transcription factors involved in the growth of mandible. Isl1 belongs to LIM- Homeodomain(LIM- HD) family of a transcription factor, playing an important role in the mature of the islet, motor neurons in the process of specialization and the development of heart. This experiment on one researches researches Isl1 gene function of mandibular growth. Our research with Shh- Cre mice with Isl1loxp/loxp mice mating, get Isl1 conditional knockout mice in the epithelial cells of the mandible. The results for the first time find that Isl1 mutant mice become lower jaw dysplasia, mandibular significantly shorter and development under the mid-line fusion, teeth and tongue defects. Through the study, we find that the jaw is regulated by the activity of the WNT and SHH pathway in the process of development, will affect many important genes that led to the development of jaw changes mandible cell proliferation and apoptosis, and finally adjust the growth of mandible. |
