Background and purpose Minimally modified low density lipoprotein(mm LDL) is a risk factor for the development of atherosclerosis, hypertension, and stroke occurrences; its mechanism is still not clear. Mm LDL is a lipid fraction oxidized LDL, can be identified by LDL receptor, and further oxidized into ox LDL. Cardiovascular and cerebrovascular diseases and vascular contractile functions are closely related, vascularsympathetic nervous system plays an important role in activity of vascular sympathetic nervous system. Sympathetic nor epinephrine released excited vascular smooth muscle alpha receptor, vascular contraction. At present, the role of mm LDL in mesenteric arteries ofalpha receptors is unclear, the experimental observation of the effect of mouse tail vein injection of mm LDL systolic function, the alpha receptor expression level of m RNA and protein. This study aims to provide theoretical guidance and experimental basis for the effect of mm LDL on stroke, new ideas for the prevention of stroke and research foundation for developing new drug.Methods 1 The preparation of mm LDL2 model and groups: mm LDL damage in mice tail vein injection of mm LDL at a dose of 1mg/kg, start time from first to q12 h after end of injection, injection of 1 times, in 96hours(i.e.in 0, 12, 24, 36, 48, 60, 72 and84 h respec tively 1injections) were sacrificed by dislocation, immediate separation contain mesenteric artery tissue, isolated mesenteric.artery andmicroscope. Respectively, in time for the units and dose units.3 DMT620 is used to measure the effect of instrument recording mmLDL on mesenteric artery systolic function of mouse vascular tension micro.4 Western Bloting detection technology is used to detect the change of expression of alpha receptor protein in control group and mm LDL group.5 The technology of RT-PCR is used to find out the expression change of m RNA alpha receptor in control group and mm LDL group. The experiment is set to discover whether mm LDL can induce the contractile function of alpha receptor.Results 1 vascular ring contraction reaction of K+ and removal of artery endothelial.2 mm LDL time and dose effect relationship between alpha receptor.NA shrink dose-response curve of mouse tail vein injection of mm LDL induced enhancement of performance for the Emax value by the NS group(120.75 + 3.44)% to(161 + 6.87) %(P < 0.01), the p EC50 value fromthe group(5.65 +0.05) up to(6.20 +0.08)(P < 0.01) alpha1 adrenocept or antagonist prazosin caused significant dose effect curve shifts to the right;3 the expression of mm LDL induced by alpha adrenoceptor m RNA levels and protein increase.Conclusions Tail intravenous injection mm LDL enhanced the vascular expressions of α1 adrenoceptors and the contractile effects mediated by α1 adrenoceptors.
