Study On Expression Of Hypothalamic Proteins Together With Points Raised By Preventingstress-induced Gastric Ulcer In Rats

Purpose: In this study, through a randomized controlled experiment in rats, using the Lower He-Froni Mu points and back-Shu and front-Mu points of acupoint selection,respectively on stress ulcer in rats by acupuncture prevention, the actual effects of different preventive measures; through the analysis of protein expression profile of SDS-PAGE protein contained in hypothalamus of rats, to further explore the effect of acupuncture the effect on expression of hypothalamic protein, protein and summarizes the acupuncture treatment of stress gastric ulcer, further to explore the preventive effect by comparing the Lower He-Froni Mu points on gastric stress ulcer. Preliminary study on the biological mechanism of acupuncture stimulation of the central methods for prevention and treatment of stress ulcer, to provide the reference for the related rules of the central "acupoints and viscera" regulation, to better serve the clinical.Method: A randomized controlled trial. 32 male SD rats, were randomly divided into ABCD. For group C respectively( "Wei shu", "Zhongwan" acupuncture group)and group D( "Housanli", "Zhongwan" acupuncture group) wereset for acupuncture treatment,use 0.5inch acupuncture needle, "Zhongwan" plug into 2mm, "Housanli" plug into 5mm, "Wei shu" plug into 5mm, adopts thereinforcing reducing techniques, once a day, in the morning 8-12 points, the needle for 15 minutes, for 10 consecutive days. B group daily crawl, were set to 15 minutes, BC and D using WRS stressmodel. A group of normal feeding, without any treatment. After the modeling, remove the four groups of rats gastric tissue and hypothalamus, the naked eye observation ofulcer index of gastric tissue, HE staining under microscope; hypothalamus after grinding, experimental SDSpolyacrylamide gel electrophoresis of grinding fluid, in order to observe the differences of protein speciesResult:1. Observation: B group showed gastric ulcer number, size is not large, the gastric ulcer index. The degree of C group and D group, the injury of gastric mucosa ulcer index of lighter,smaller, and the he Mu points group is better than the Yu Mupei point group, more close to the blank control group.2. HE staining: Microscope was used to observe the gastric mucosa layer structure of A group rats, epithelial cells and glands arranged neatly, no capillary dilatation and congestion,no inflammatory cell infiltration. B group, located in gastric mucous layer visible ulcer, local mucosal layer interruption, and shedding of epithelial cells, glands were damaged to someextent, are seen in the submucosa congestion edema and inflammatory cell infiltration. C group and D group rats gastric mucosa layer, the integrity of epithelial cells and glands, the arrangement of capillaries and inflammatory cell infiltration in aspects of performance are good. Moreover, there was no D group. Cells, cells arranged in neat rows, no inflammatory cell infiltration, the rat gastric mucosal morphology compared with the C group to A group.3. SDS- polyacrylamide gel electrophoresis :There was significant difference between the contents of different protein of the four groups of rats in the hypothalamus, the relative molecular weight of protein band near 72 k Da,43 k Da,34 k Da, B group were significantly higher than that of A group, C group and D group after acupuncture prevention, and its content was significantly lower than that of B group, and D group compared with C group,, its content is more close to the A group; the relative molecular weight of protein band near95 k Da, B group was significantly lower than that of A group, C group and D group after acupuncture prevention, and its content was higher than that in group B, and group D,compared with C group, the content is more close to the A group.Conclusion: Acupuncture therapy on the curative effect of prevention and treatment of stress ulcer is exact,and the Lower He-Froni Mu points compared with the back-Shu and front-Mu points has more significant clinical effect. To speculate its mechanisms, relative molecular weight about 95 kda protein secretion levels were lower and the relative molecular weight is about 72 k Da, 43 k Da, 34 k Da protein secretion is closely related to the incidence and stress gastric ulcer, acupuncture is likely to through to change this trend, so as to prevent the occurrence of stress gastric ulcer, and raised with the acupuncture group was inhibitory effect was better than that of Shu Mu acupuncture point group. Which relative molecular weight is about 95 kda protein may be as the heat shock protein HSP90-α(hsp90aa1), 72 k Da protein may be homologous 71 k Da heat shock protein(hspa8) and 70 k Da related heat shock protein(hspa2), 43 k Da and 34 k Da protein may for the enol enzymes(enolase), They are secreted by the hypothalamus or the secretion of hypothalamus after he transferred to the transfer or action. The chemical nature of the secretion and transfer process, further research should be done.
