ObjectiveStudy on material basis of Sanhuang Decoction extract for the Preventive effect of hemolytic disease of newborn.Methods1.Identified each medicine related the components of Sanhuang decoction(SHD) in source, character, microscopic identification by reference to Chinese medicine Yinpian identification project of the 2010 version of "Chinese Pharmacopoeia".In order to guarantee the reliability and efficacy of the study of pharmacodynamic material basis,to do the quality of the medicinal quality detection.2.To identify the extract of Sanhuang decoction by microemulsion thin layer chromatography(METLC).To isolated and identify important components of compound extract which complete the specificity identification of each medicine to guarantee quality of self-made compound preparation from the perspective of qualitative.3.To establish the fingerprint of extract of Sanhuang Decoction by RP-HPLC method and the patterns of the common peaks were attributed by medicinal material source.Meanwhile,according to the data information which from the fingerprint,the 10 batch of self-made Sanhuang Decoction extract was conducted to evaluate the quality by using chemometrics methods to guarantee the quality of self-made compound preparation from a quantitative point.4.To establish the Serum fingerprint of rats which were orally given Sanhuang Decoction extract,and compared with the date of vitro fingerprint to determine the source, type and quantity of the blood transitional components of Sanhuang Decoction.Changes of the relative content of each component was observed, with reference to the relevant literature and according to the serum pharmacochemistry(SPC)of relevant theoretical screened the efficacy of material basis of Sanhuang Decoction.5.To establish the animal model of blood group incompatibility hemolytic disease from blood group antigen antibody humoral immunology by using indirect method.A preliminary study of animal experiment was carried out on effects of Sanhuang Decoction extract and its possible pharmacodynamic basis material 6,7-dimethoxycoumarin in the treatment.Through the comparative efficacy to determine the position of 6,7-dimethoxycoumarin as the pharmacodynamic material basis of Sanhuang Decoction for prevention of hemolytic disease of newborn.Resμlts1.The resμlts of qualitative identification of Herba Artemisiae, Radix Scutellariae,Cortex Phellodendri, Radix Glycyrrhizae,Radix Rhizoma Rhei were showed that the above Chinese medicine Yinpian are regular source of high-quality medicinal materials.2.The microemμlsion TLC of compound extract,through a sample,various main components including Chlorogenic acid, berberine hydrochloride, baicalin, rhein, emodin, chrysophanol and other ingredients from Sanhuang Decoction extract can be effectively separated out simultaneously in the same piece of thin layer plate,and the spots are round and set, clearly visible, no tail phenomenon.3.Sanhuang Decoction in vitro chemical composition can be expressed by a fingerprint, and the method is accurate,reliable,and reproducible.The similarity of 10 batches of samples were above 0.9, and the principal component analysis and cluster analysis results were generally consistent, further verified the stability of prescription preparation process consistency and quality control of the preparation.These guarantee the effectiveness of study on the compound prescription pharmacodynamic material basis.4.Fingerprint of serum components of Sanhuang Decoction extract on rats after intragastric administration with different time points was established, to determine the source, type and quantity of Sanhuang Decoction blood transitional components.According to relative change of the composition and the result of related literature reference, preliminarily determines the 6,7-dimethoxycoumarin or the compounds as active ingredients of Sanhuang Decoction for the effects of the extract to prevent hemolysis disease of the newborn.5.The animal model of blood group incompatibility hemolytic disease was established indirectly from the perspective of the humoral immunological.When compound compared with monomer effect, Sanhuang Decoction extracts and 6, 7-dimethoxy coumarin can reduce ABO blood group antibody titer, and the effects of the monomer was more obvious than compound under the condition of selected best dosing. This research helps to reveal the immune function of Sanhuang Decoction in treat the hemolytic disease of newborn,and provides the reference for further study of the mechanismof reduce Ig G antibody titer of Sanhuang Decoction.Conclusions6,7-dimethoxy coumarin as monomer extracted from Sanhuang Decoction can effectively reduce ABO blood group antibody titer of the immune rats.It is the pharmacodynamic material basis of Sanhuang Decoction for prevention of hemolytic disease of newborn.
