| ObjectiveThe fruit of Celastrus Orbiculatus was one ingredient of Yixinningshen Tablets. New Yixinningshen Tablets was the fruit of Celastrus Orbiculatus replaced by Albizia Julibrissin. Albizia Julibrissin had a good effect on insomnia, many depression prescription had Albizia Julibrissin. Studies had shown that flavonoids was the highest and main active ingredient of Albizia Julibrissin. In order to further study the New Yixinningshen Tablets, we optimized the extraction technology of total flavonoids from Albizia Julibrissin. New Yixinningshen Tablets and Yixinningshen Tablets were compared the efficacy strength by HE staining and immunohistochemical staining, and explored their mechanism of action. We studied on sedative and hypnotic effects of the volatile oil from New Yixinningshen Tablets. A method was established for simultaneous determination volatile components and provided a reference for the further quality control of New Yixinningshen Tablets.MethodsThe surfactant concentration, liquid to material ratio and ultrasound time were investigated by response surface methodology on the effects of extraction rate to determine the optimum extraction conditions of total flavonoids. Healthy adult rats were randomly divided into New Yixinningshen Tablets prescription one, two, three, four groups, Yixinningshen Tablets group, Zaoren sedative particle group, sleep deprivation group and the normal group, in addition to the normal group, the remaining rats were sleep deprivation, the nerve cells morphology of rat hippocampal dentate gyrus were observed by HE staining, 5-HT and GABA were stained by immunohistochemistry three step method. We analyzed the mean optical density value size by Image Pro Plus. We gave the volatile to mice and analyzed the effect on the central nervous system accord to the locomotor activity experiments. The bornyl acetate, piperonyl alcohol and caryophyllene were simultaneous determination by GC.ResultsSDS concentration was 2.5%, material liquid was15 times, ultrasonic time was 31 min. Under these conditions, the extraction rate of total flavonoids from Albizia Julibrissin was the highest, reaching to 0.853%. Using this technology, the extraction yield increased by 0.411% as compared with that using ultrasonic technology alone. To observe the structure changes of nerve cells under the light microscopy, each treatment group was significantly less than the sleep deprivation group, New Yixinningshen Tablets prescription two groups rather less than Yixinningshen Tablets group, the results of immunohistochemistry showed that compare to sleep deprivation group, the expression of 5-HT and GABA were significantly increased, the expression of New Yixinningshen Tablets prescription two groups more than Yixinningshen Tablets group, and the difference was significant. The low, medium and high dose of volatile oil had significantly inhibited of locomotor activity in mice. The average recoveries(n=3)of bornyl acetate, piperonyl alcohol and caryophyllene were 99.06%, 99.34%, 98.72%. RSD were 0.53%, 0.22%, 0.70%.ConclusionsThe optimum extraction technologies can be used for extraction of total flavonoids from Albizia Julibrissin for its simplicity, feasibly and credibly, the technologies can be used to provide a scientific basis for extraction of total flavonoids from Albizia Julibrissin and New Yixinningshen Tablets. Each treatment group had significant therapeutic effect to the injury of rat hippocampal dentate gyrus caused by sleep deprivation, New Yixinningshen Tablets prescription two groups rather stronger than the Yixinningshen Tablets group, its mechanism of action may be related to the protection of cell normal structures and increased the expression of 5-HT and GABA. The volatile oil had sedative effect on mice, was one of the main active ingredient of New Yixinningshen Tablets. A method was established to simultaneous determination bornyl acetate, piperonyl alcohol and caryophyllene in New Yixinningshen Tablets by GC. The method was accurate and reliable, can be used to control the quality of New Yixinningshen Tablets. |
