| Objective To analyze the correlation between plasma mi RNA-32 level and coronary artery calcification, and to investigate the relationship between mi RNA-32,ALP and Runx2 during the progression of coronary artery calcification, providing experimental basis for the utilization of mi RNA-32 as molecular marker for the diagnosis and prognosis of coronary artery calcification.Method(1) The CAD patients with CAC were determined by and CT scanning(Siemens Definition Flash, Siemens Healthcare, Forchheim, Germany) and patients are divided into CAC group( calcification group,including 33 samples) and non CAC group(control group, including 33 samples)(2) Q-PCR was used to detect the plasma mi RNA-32 level; the ALP activity and Runx2 level in patient plasma were determined by enzyme activity assay and ELISA assay respectively.;(3) Correlations between plasma mi RNA-32,ALP and Runx2 were analyzed by pearman’s rank correlation coefficient analysis.Results It was revealed that mi RNA-32 level showed a markedly increase in plasma of CAD patients with CAC compared to those without CAC(p=0.016). When further stratifying the patients according to the age, we found that plasma mi RNA-32 presented a higher level in patients with CAC compared to those in patients without CAC at the ages from 50 to 59(p=0.027). It was also found that plasma mi RNA-32 showed an significant increase in male patients with CAC compared to those without CAC(Figure 6E)(p=0.013).It was unveiled that plasma ALP show a higher level in CAD patients with CAC compared to those without CAC(Figure 6F)(p=0.026) and we observed a positive correlation between the plasma levels of mi RNA-32 and ALP in CAD patients at the ages of 50 to 80 with CAC(r=0.547, p=0.001),Conclusion Plasma mi RNA-32 was revealed to show an increased level in CAD patients with CAC compared to those without CAC and a positive correlation between the plasma levels of mi RNA-32 and ALP in CAD patients was found, which indicate that mi RNA-32 might serve as a diagnosis molecule for the CAD patients with CAC,... |
