Objective:To detect the positive expression rate of PNL2 in malignant melanoma tissueand and to evaluate the clinic significance and its relationship with melanoma diagnose nvasion,metastasis,prognosis and BRAF mutations.Methods: Immunohistochemistry was carried out to quantify the expression of PNL2 in paraffin-embedded tissue samples from 80 cases of MM.The relation between PNL2 expression and Clinical pathological features was analyzed in these lesions.Test results chi-square four tables exact probability method is used to analyze PNL2 in different age, gender, nationality, lymph node metastasis, Clark, grading, infiltrating depth and prognosis, the differences in the expression of melanoma.Results:Immunohistochemistry revealed that the expression rate of PNL2 was 91.25%(73/80) in melanoma tissue. The PNL2 expression was positively correlated with lymph node metastasis, Clark grade, depth of invasion,prognosis(P<0.05)5 but uncorrelated with the Age,gender and ethnicity (P>0.05).15 cases were positive for PNL2 expression occurred in 19 cases BRAF mutant melanoma tissues,including 14 cases were positive for PNL2 expression occurred in 15 cases of melanoma of BRAFV600E mutation. Conclusions:PNL2 is a highly sensitive and specific melanoma marker that is useful to the clinical diagnosis and prognosis of melanoma. This study prompt that PNL2 appears to be a valuable supplementary reagent for the diagnosis of melanocytic tumors.The positive expression of PNL2 in BRAFV600E mutation melanoma provides a theoretical basis for looking PNL2 specific antigen.
