Rehmannia glutinosa Libosch belongs to the genus Rehmannia of Scrophulariaceae, Mainly distributed in Henan, Shanxi and other provinces in China. The roots of this plant have been used as an anti-pyretic, an anti-anemic and a tonic. The chemical constituents of Rehmannia glutinosa include saccharides, iridoids and phenylethanoid. In previous work, the chemical constituents of the aerial parts were similar to the root of Rehmannia glutinosa, which prove that the aerial parts also had high value of medicinal purposes. However, the aerial parts of Rehmannia glutinosa are underused. Study on chemical constituents of the aerial parts of Rehmannia glutinosa not only could enhance utilization of rehmannia resources, but also could find new active ingredient resources.Twenty-six compounds were isolated by macroporous, resin silica gel, Sephadex LH-20 and HPLC chromatographies from the aerial parts of Rehmannia glutinosa. Fourteen compounds were elucidated on the basis of spectral analysis (MS, 1D NMR and 2D NMR). The structures of the isolates were identified names as (+)-(7’S,8S, 8’S)-9’-O-[β-glucopyranoside] sesaminone(1),3-hydroxy-4-(4-(2-hydroxyethyl) phenoxy) benzaldehyde(2), glutinosalactone A(3),2α,3β,19α,23-tetrahydroxyolean-12-en-28-oic acid(4), chrysoeriol(5), apigenin(6), luteolin(7),7.3’-dihydroxyl-5’methoxyisoflavone(8), aeginetic acid(9), cinnamic acid(10), isovanillic acid(11), pyrocatechol(12), corchorifattty acid B(13) and pinellic acid(14) respectively. Among the isolated compounds 1 and 2 were new compounds and compound 4,8,13 were isolated from this genus for the first time.The MTT assay was used for quantitative determination of anti-tumor activities. Compound 7 exhibited moderate cytotoxicity against HCT-8 cell. Others compounds did not show cytotoxicity against the HCT-8 and BEL-7402 cell.
