Objective:Analyze the different expression of miR-146a\miR-133b in cervical tissues and the gene polymorphism of miR-146a (rs2910164) in Xinjiang Uyghur and Han nationality, explore the relationship between miR-146a\miR-133b and the occurrence and development of cervical cancer. Methods:The PCR-RFLP to detect the genotype of miR-146a (rs2910164) in the paraffin embedding tissue of cervicitis, cervical intraepithelial neoplasias and cervical cancer. The RT-qPCR to detect the relative expression of miR-146a\miR-133b in paraffin embedding tissues of cervicitis, cervical intraepithelial neoplasias and cervical cancer. Results:The result of PCR-RFLP shows the significant difference between Uyghur and Han in constituent ratio of miR-146a (rs2910164) genotype. Uyghur women carry allele G proportion (64.55%) were significantly higher than that of the nan nationality (41.80%). The Logistic regression analysis of miR-146a(rs2910164) genotype and clinicopathological characteristics found that the Uyghur (OR=3.332,95% CI:1.411-7.868) and tumor diameteris was greater or equal than 4cm (OR=3.792,95% CI:1.226-12.376) were significantly associated with GG\CG genotypes. Compared with cervicitis, miR-146a\miR-133b increased the expression in CIN and cervical cancer significantly. With the cervical lesion is aggravating, the expression level increased. The miR-146a (rs2910164) GG genotype expression level was significantly higher than CC genotype in CIN and cervical cancer tissues (P=0.019).If marriage age less than 20 years old, tumor diameter was greater or equal than 4cm, with HPV infection, miR-146a\miR-133b was in a significant higher expression. Conclusions:MiR-146a (rs2910164) gene polymorphism was significantly associated with national factor and tumor diameter.MiR-146a (rs2910164) allele G associated with the high expression of miR-146a, may participate in the progression of cervical cancer. MiR-146a\miR-133b significantly increased expression level in the cervical lesion, may become a new molecular markers of prediction and evaluation in cervical cancer.
