Experimental Study On Chen Yuangao Ointment Massage On KOA Rat Cartilage Matrix And TNF-α And TGF-β1 Effects

Objective:To explore the mechanism of action and clinical significant of Chenyuan Ointment for controlling and treating knee osteoarthritis,and to provide the theory evidence for the clinical application.Methods:Selected 52 apinoid SD rats and divided the 52 SD rats into 4 groups randomly:normal group (group A), model group (group B), Calligraphy control group(group C) and Chenyuan cream group (group D). Fixed every rat’left knee-joint of B、C、D group by closed-plaster method 8 weeks to induce the KOA model. After 8 weeks fixation, the rats in A、B group without intervention, the rats in group C、D group were severally given Chenyuan cream with manipulation and Vaseline with manipulation once a day. After 8 weeks, detected the serum levels of TNF-a and TGF-β，HE staining, observed the pathological changes of articular cartilage in each groups and Mankin scoree. Results:Compared with B group, C、D group showed lower Mankin score and the serum levels of TNF-a,Between C group and D group,D group showed the lower serum levels of TNF-a(P<0.05);Compared with B group, C、D group showed higher the serum levels of TGF-β,Between C group and D group,D group showed the higher serum levels of TGF-β(P<0.05);Compared with the model group, two rats in the treatment group after treatment, the lower limb circumference and activity of knee joint were increased significantly (P<0.01), in which Chen Yuan paste test group increased by a big margin (P<0.05)..Conclusion:Destruction of lesion joint friction and Chen Yuangao method can reduce the KOA of articular cartilage, the possible mechanism is through the destruction of inhibition of inflammatory cytokines TNF-alpha, promote the transformation growth factor beta 1 TGF-repair function, improve rat lower limb muscle strength and knee joint activities to achieve.
