Objective1. To analyze the expression and prognostic impact of CREPT on retroperitoneal liposarcoma.2. To analyze the effect of CREPT on SW872.Methods1.The records of 101 patients with retroperitoneal liposarcoma managed with surgery between June 2007 and March 2014 were retrospectively reviewed. HE staining and immunohistochemical staining was used to evaluate the impact of CREPT on retroperitoneal liposarcoma.2. Gene silencing was made by RNA interference (RNAi) technology and the effect of CREPT on the proliferation of SW872 was analyzed by MTT method, cell plate cloning experiments and scratch test.ResultsCREPT was high expressed in retroperitoneal liposarcoma and was significantly related to tumor grade (x2=67.070, P<0.01), mitosis (x2=52.055, P<0.01), tumor necrosis(x2=25.546, P<0.01) and tumor differentiation(x2=25.546, P<0.01). Univariate analysis showed expression of CREPT (x2=37.759, P<0.01), tumor grade, mitosis (χ2=71.644, P<0.01), tumor necrosis (x2=27.369, P<0.01), tumor differentiation (χ2=30.357, P<0.01) were positively correlated with survival. Expression of CREPT (RR=2.289,P=0.015), mitotic (RR=2.444,P=0.005) were independent prognostic indicators for survival. CREPT can enhance the proliferation of SW872 and gene silence of CREPT can decrease the ability of proliferation through MTT method, cell plate cloning experiments and scratch test (P<0.05)ConclusionsCREPT in retroperitoneal liposarcoma has a high expression, and it was significantly related to prognosis. Cell proliferation of SW872 could be decreased by gene silencing of CREPT.
