Currently, due to difference of the price between Lonicerae japonicerae flos (LJF) and Lonicerae flos (LF), there is a phenomenon that the two species mixed used on the market. But their clinical effects are different. So it is necessary to set up the methods of identification and quality control of two kinds of traditional Chinese medicines.Based on this, in combination with the current domestic and international research progress of traditional Chinese medicine (TCM) in quality control and fingerprint, this article explored the quality control methods of LJF and LF from three aspects respectively:rapid identification, simultaneous determination of content of multi index component, and the fingerprint establishment and comparison research.The main work and achievements are as follows:1. Through consulting a large number of literatures, this paper makes the summary of the domestic and foreign developments, the prospects and limitations of the TCM fingerprint. At the same time, a review of distinguishes between LJF and LF and their present situation of quality research are given.2. Taking the macranthoidin B and asperosaponin B as the distinctive constituents, an UPLC-ELSD method for the rapid identification of the authenticity of the materials was established. And the feasibility of this method was proved through the investigation of 13 batches of LJF and 14 batches of LF.3. An UPLC-PDA method for the simultaneous determination of the content of 9 active constituents, including organic acids (chlorogenic acid, caffeic acid, neochlorogenic acid, cryptochlorogenic acid, isochlorogenic acid A, isochlorogenic acid B, isochlorogenic acid C) and flavonoids (Luteolin-7-O-glucoside, rutoside) in LJF and LF was established.4. Fingerprints of LJF and LF were established respectively by UPLC-UV method, as used to determine the LJF 40 common peaks and LF 15 common peaks. With the combination of "chromatographic fingerprint evaluation system 2004 edition", quality evaluation of LJF and LF were set up on their own reference fingerprint by the similarity results (qualified or unqualified). Comparatively studying on the composition for the two kinds of materials, the research found that the number of chemical composition containing in LF is obviously less than that in LJF.14 out of 15 characteristic peaks in LF were shared with the later, and of which seven common components were calibrated by the retention time of reference substances:including neochlorogenic acid, chlorogenic acid, caffeic acid, cryptochlorogenic acid, isochlorogenic acid B, isochlorogenic acid A, isochlorogenic acid C. While in the special components of LJF, Luteolin-7-O-glucoside and rutoside are marked. And the study found that the peak at the relative retention time RRt=2.310), in which chlorogenic acid as reference peak, has a larger height and area, which is more significant to be used to distinguish the two species compared with the regulation of Pharmacopoeia where Luteolin-7-O-glucoside is used to be the distinctive feature.In conclusion, this subject made a systematic study on quality control of two species and carried out the research of the feasibility of the traditional Chinese medicine (TCM) fingerprint, which has laid the foundation for the rational development and utilization of resources of Lonicerae japonicae flos and Lonicerae flos.
