Objective:○1 Based on the last 10 years in our hospital inpatients of asthma by the retrospective investigation,we analysis TCM syndrome distribution regularity of asthma,and provide the TCM clinical syndrome treatment basis for the development and optimization of asthma.○2 To learn the pathogenesis of asthmatic rat airway remodeling models,observe the effect of Mashaopingchuan decoction on asthma models for male SD rats morphology,lung function,research on the mechanism of action of Mashaopingchuan decoction prevention and treatment of asthma from cytokine and airway remodeling, and provide experimental preclinical for Mashaopingchuan decoction about clinical application.Methods:○1 With relevant literatures, make the table of retrospective questionnaire of asthma. To investigating 261 cases of patients with asthma in this study, statistically analysis the relevant factors in order to explore the TCM syndrome distribution characteristic of asthma, with the relationship between general information(gender, age,occupation, duration, etc.) and predisposing factors(history of tobacco and alcohol,allergies, genetic history, climate change, food fatigue, etc.) and asthma among TCM syndromes.○2 The 70 male SD rats were randomly divided into normal group, model group, Mashaopingchuan decoction low, medium and high dose group, dexamethasone group and Yang He Asthma-Easing group, each group of ten only. Except the normal group, the other groups in the experiment on the day of 0th, 7th and 14 th were all used10% ovalbumin hydrotreating 10 mg aluminum oxide powder. Multi-point injection sensitization between intraperitoneal and both sides of the groin skin, then preparation of asthmatic rats with inhalation inspiring; The intervention group were administered beginning from the 7th day of the challenge phase, gavaged 30 min before each excitation, and normal group and the model group with normal saline instead in 14 consecutive days; intraperitoneal anesthesia, tracheotomy and line spirometry wereconducted 24 hours after the last administration. The changes of lung tissue pathology morphology was measured by HE; analysised of morphological parameters of the airway and measured the degree of airway remodeling with computer image analysis software; determination of lung tissue TGF-β1, PDGF-BB content with immunohistochemistry; serumed IL-5, IL-13 content with ELISA; the gene level of TGF-β1 and α-SMA in rats lung tissue was measured by RT-PCR;the protein level of TGF-β1 and PDGF-BB in rats lung tissue was measured by Western blot.Statistical analyse the results by SPASS software.Results : ○1 In 261 cases of asthma, cold whooping 122 cases(46.7%), the most common, followed by 87 cases of hot whooping, accounting for 33.3%. 112 cases were male(42.9%), female 149 cases, accounting for 57.1%, the incidence of women than men, and illustrates the onset of asthma often witness and type symptoms. ○2 Each group has different degrees of airway remodeling, Mashaopingchuan high dose group of rats degree of airway remodeling(bronchial wall thickness, muscle thickness) was significantly lighter than the dexamethasone group(P <0.05); Mashaopingchuan high dose group and Yang He Asthma-Easing group of lung function and rats pulmonary function FVC,FEV0.3,PEF were improved most significantly(P<0.05),followed by dexamethasone group; Mashaopingchuan decoction could reduce the expression of serum IL-5 and IL-13(P<0.05); Mashaopingchuan high dose group、 dexamethasone group and Yang He Asthma-Easing group could reduce the content of TGF-β1,PDGF-BB and α-SMA.Conclusions:○1 From 261 syndrome asthma, cold whooping is the most common type,the onset of symptoms of the common white phlegm cough, aversion to cold and fever,wheezing, chest tightness, no chest pain, no thirsting, no sweating, greasy tongue coating, floating pulse tight.○2 In a rat model of asthma, Mashaopingchuan decoction could reduce airway inflammation, reversing airway remodeling, the mechanism may beby reducing IL-5 and IL-13 content, inhibit the expression of TGF-β1, PDGF-BB andα-SMA, indicates TGF-β1, PDGF-BB and α-SMA is closely related to the degree of airway remodeling, may be used as one of the measure of severity of airway remodeling index.
