Background:Sepsis is infection that caused by all kinds of pathogenic microorganisms or their toxins in the body’s blood or tissue. It also can cause Systemic inflammatory response syndrome (SIRS). Sepsis is one of the most important symptoms which is harmful to human, there are about 18000000 people suffering from sepsis in the world each year, about 750000 people were diagnosed only America. And sepsis is one of the reasons to death. For example, in America, sepsis is eighth of death reason in 65 to 75 years old of age. The patients with sepsis is increasing due to the aging of the population, abuse of antibiotic and hormone, increase of the transplantation operation, increases of patients with diabetes, the rising prevalence of tumors due to the use of immunosuppressive agents. Early prevention and treatment of acute kidney injury induced by sepsis, has very important significance. In recent years, thymic peptide five (Thymopentin-5, TP-5) is widely used in severe burns, immune regulation of chronic hepatitis. This study establish the model of acute kidney injury in septic rats by cecal ligation and puncture (CLP) to investigate the effect of TP-5 on inflammatory response and immune function in rats with sepsis and to provide evidence for clinical rational use.Objective:Establish the model of acute kidney injury in septic rats and investigate the effect of TP-5 on inflammatory response and immune function in rats with sepsis.The further discussion of thymopentin-5 on protective effect and mechanism in rats with sepsis caused acute kidney injury(AKI).Methods:90 cases of healthy and male SD rats (weight230-250g) were randomly divided into 3 groups:control group(C group), sepsis group (S group), thymopentin group(T group).These groups were divided into five time point including lh,6h,12h,24hand 48h with 6 rats in each time point. The sepsis model was made by CLP. The blood and kidney tissue were collected in each time point. Changes of renal pathology were observed under light microscope and relevant indexes like serum creatinin(Cr), blood urea nitrogen(Bun), tumor necrosis factor a(TNF-a), interleukin-10(IL-10), CD4+/CD8+ was tested and analyzed. Using of light microscope to observe the pathological changes of kidney tissue.Results:Pathological concentration in three groups of rats kidney. The Bun comparison in three groups of rats. The Cr concentration in three groups of rats. The TNF-a concentration in three groups of rats. The IL-10 concentration in three groups of rats. The CD4+/CD8+concentration in three groups of rats.In T group, concentrations of Cr, Bun 6h after CLP starts to rise,reached peak in 24h and decreased in 48h,which were all lower than S group(P<0.05) and higer than C group(P<0.05).Compared with C group, concentrations of TNF-a 1h significantly rise,reached peak in 12h and decreased in 24h, which were all lower than S group(P<0.05) and concentrations of IL-10 1h significantly decreased, reached peak in 12h and rise in 24h, which were all more than S group(P<0.05). In T group, CD4+/CD8+ ratio 6h after CLP starts to decreased reached the lowest in 24h and rise in 48h,which were all lower than S group(P<0.05) and significantly lower than C group(P<0.05).Conclusion:1. Sepsis-induced acute kidney injury animal models were made by cecal ligation puncture (CLP).2. Thymopentin-5 treatment can alleviate limb ischemia reperfusion acute kidney injury,it plays the role of renal protection in AKI caused by sepsis.
