Typing of TCM syndrome type of avascular Osteonecrosis of Femoral Head has been the lack of strong and direct evidence to confirm, difficult to achieve the objective of the real meaning and quantization, and provide scientific basis for typing to study the TCM syndrome type of femoral head necrosis. The phenotype of chondrocytes of different syndromes in different, through the research on MAPKs signal differences, this study hopes to look the part of the solution to the essence of TCM syndrome type of avascular necrosis of the femoral head location.ObjectiveThe establishment of different syndromes of patients with osteonecrosis of the femoral head femoral cartilage cell in vitro culture system, using the technique of genomics and proteomics,to investigate the MAPKs signal differences of different syndromes of chondrocytes, dig out the common features associated with asyndrome markers, provide objective and quantitative basis for research on theessence and reasonableclassification of disease syndromes of traditional Chinese medicineavascular necrosis of femoral head.MethodsThe collection of avascular necrosis of the femoral head in patients with a total of 20cases, and in accordance with the blood stasis type, liver and kidney deficiency type of syndrome differentiation and packet. Patients undergoing total hip replacement, the collection of different syndromes of intraoperative cartilage, were culturedchondrocytes in vitro, detection and value-added and apoptosis. Changes of different syndrome analysis of MMP-1/-13mRNA type of articular cartilage cells usingreal-time quantitative PCR and Western blot method to detect MMP-1 and MMP-13expression of mRNA in different syndrome types and protein, and the establishment of the corresponding protein atlas. In mass spectrometry shows are non consistent protein.ConelusionThe experimental results suggest that the apparent gap, and provides a new idea for the further study of typing of TCM syndrome type of femoral head necrosis, andbecome a large sample of experimental research is carried out according to the "preexperiment".
