ObjectiveExtracting and separating the distinctive chemical composition from the dried fruits of Syzygii aromaticum,and establishing a good quality standard which can provide references for the quality control.The fruit of Syzygii aromaticum was used for spleen and stomach diseases, vomiting,diarrhea and other diseases. It is also one of the famous spice and traditional Chinese medicine which is planted in Tanzania, Sri Lanka, Madagascar, Indonesia etc. Here in China, it is also planted in provinces like Hainan, Guangdong, Guangxi etc.According to the ancient medicine books, Syzygii Fructus and Syzygii Flower buds share a lot in common functionally but also have their own differences. For their growth periods are distinct which leads to different effects.According to the Chinese Pharmacopoeia which published in 2010, the inspection projects of Syzygii Fructus including character, microscopic identification, TLC of eugenol, moisture, total ash, extract, determination of eugenol. And the current inspections are quite simplex and short of specificity, somehow similar with the inspections of the flower buds of Syzygii aromaticum.It’s necessary to analyze the chemical content of Syzygii Fructus and establish a good quality standard.MethodsIt has been reported that the extracted chemical component from Syzygii Fructus are mainly the volatile oil and a few of phenylpropanoids, flavonoid, triterpenoid etc. This thesis is to study the petroleum ether extraction from the methanol extraction of Syzygii Fructus. Extractding the eugenol and bancroftione these two reference substances by the silica gel and ODS column chromatographs, and use TLC to detect them. According to formulary requirement, we use the TLC method and HPLC to identified its purity, and identify the structure by spectral analysis and physical chemical data.The research established TLC method of eugenol and bancroftione, and the content of the two were determined by HPLC. Meanwhile, we established a specific chromatogram of Syzygii Fructus by GC, which is different from Syzygii Flower buds. Identifying the structure and calculating the relative content of seven Gas Chromatography groups by using the Agilent MassHunter Qualitative AnalysisB.06.00 MS workstation Service and the NIST MS Search 2.0 mass spectral library. Moreover, utilizing the inductively coupled plasma mass spectrometry to determine whether the contents of Pb, Cd, As, Hg and Cu in all of 10 samples were within the limitation or not. By applying the HPCEC method to determine the content of aflatoxin, all these will provide reference for the security application of Syzygii Fructus. In addition, reviewing other inspections of original standard was reasonable or not.ResultsThe experiment extracted and purified the referential substances of bancroftione from the fruit of Syzygii aromaticum. And it improved the TLC method of Syzygii Fructus by adding the bancroftione as its control article. Meanwhile, it controlled the content of eugenol and bancroftione. It established the specific chromatograms of Syzygii Fructus also for the first time. Seven peaks were defined, they were alpha.-Cubebene, Copaene, Caryophyllene, Cyclohexane,1-ethenyl-1-methyl-2-(1-methylethenyl)-4-(1-methylethylidene), Naphthalene,1,2,3,5,6,8a-hexahydro-4,7-dimethyl-1-(1-methylethyl), 1S-cis, 2-hydroxy-4,6-dimethoxy-5-methylacetophenone. Similarity Evaluation System for Chromatographic Fingerprint of TCM (2009) was used to analyze the 10 batches of Syzygii Fructus..which showed the similarity of fingerprints was more than 0.98. As opposed to eugenol,the relative retention time of each peaks should be around±5% of specified value. The specified value were 0.45(Peak No.1),0.48 (Peak No.2)、0.57 (Peak No.3)、 0.63 (Peak No.4)、0.71 (Peak No.5),1.00 (Peak No.6)、1.15 (Peak No.7),the ratio of Peak No.6 and Peak No.7 is greater than 0.1. In addition, the research studied the 5 heavy metals, and aflatoxin for the first time which provides the reference of security application of Syzygii Fructus. After reviewing the extraction of Syzygii Fructus, this item was not reasonable, so suggesting removing it from the new standard.ConelusionThis paper aims to build up the quality standard of Syzygii Fructus through systematic research, including the character, microscopic identification, TLC, Content Determination, characteristic chromatograms and so on which can control the quality of Syzygii Fructus effectively.
