| Purpose:We examined the effect of EGCG on the invasion and expressions of MMP-2, MMP-9 and VEGF in gastric cancer cells, so as to further clarify the mechanism of gastric cancer invasion. The data was generated by the study will be invaluable on the control of early gastric cancer invasion and could offer potential therapeutic targets.Methods:The gastric cancer cells (MKN-45, SGC-7901) were cultured in the absence or presence of various concentrations of EGCG (0,3.13,6.25,12.5,25,50,100,200 μM) for 24h of 37 ℃. CCK-8 was used to measure the rate of growth inhibition by EGCG in gastric cancer cell and then select the appropriate concentration as the working concentration of our subsequent experiments. Transwell invasion experiment was used to measure the invasive ability of gastric cancer cells in different concentrations of EGCG for 24h of 37℃. qRT-PCR was used to measure the MMP-2, MMP-9 and VEGF expressions of gastric cancer cells.Results:1. In MKN-45 cells, in response to EGCG of 0,3.13,6.25,12.5,25,50,100,200μM, the growth inhibition rates were 0%,11.1%,15.2%,21.1%,36.4%,48.4%,59.4%,63.2%. In SGC-7901cells, in response to EGCG of 0,3.13,6.25,12.5,25,50,100,200μM, the growth inhibition rates were 0.0%,7.3%,12.5%,19.2%,29.6%,35.7%,52.9%,59.4%. The IC50 of EGCG was 67.03μM in MKN-45 and 100.94μM in SGC-7901.2. By transwell invasion assay treatment of MKN-45 cells and SGC-7901 with EGCG (67.03μM for MKN-45 and 100.94μM for SGC-7901), significantly in inhibite their invasion rate compared to controls (MKN-45:40.24 ± 5.78% vs 100.00 ± 12.34%,SGC-7901:50.56 ± 8.13% vs 100.00 ± 19.47%, P<0.05).3. qRT-PCR assay was used to measure the MMP-2, MMP-9, VEGF expressions of gastric cancer cells in different concentrations of EGCG line 24h under the temperature of 37℃. The expressions of MMP-9, MMP-2, VEGF mRNA in the MKN-45 experimental group than the control group are:0.45 ± 0.04,0.31 ± 0.02,0.39 ± 0.02. The expression of MMP-9, MMP-2, VEGF mRNA in the control group are:1.00 ± 0.03,1.00 ± 0.05,1.00 ± 0.07. The experimental group is decreased significantly relative to the control group, P<0.5, with significant difference. The expression of MMP-9, MMP-2, VEGF mRNA in the SGC-7901 experimental group than the control group are:0.42 ± 0.03,0.30 ± 0.07,0.25 ± 0.06. The expression of MMP-9, MMP-2, VEGF mRNA in the control group are:1.00 ± 0.05,1.00 ± 0.08,1.00 ± 0.07. The experimental group is decreased significantly relative to the control group, P<0.5, with significant difference.Conclusions:EGCG can inhibit the invasive ability of gastric cancer cells.The possible mechanism could include the decreased expressions of MMP-9, MMP-2 and VEGF. |
