Objective: To investigate the expression of enhancer of zeste homolog 2(EZH2) and p53 in breast adenosis tissues and breast cancer, and analyze the relationship between two expression and the clinical pathologic characteristics and prognosis of breast cancer patients. Study the role of EZH2 and p53 in the development of breast cancer and their possible implications. Methods: The expression of EZH2 and p53 protein were detected by immunohistochemical(IHC) method in 50 cases of breast adenosis tissues, 95 cases of breast invasive lobular carcinoma(ILC) and 400 cases of breast invasive carcinoma of no specific type(NST), and analyzed the correlation between them. Results: The positive rate of EZH2 protein in ILC, NST and breast adenosis tissues were 53.7%(51/95), 51.5%(206/400), 24.0%(12/50). There was no statistical significance of EZH2 between ILC and NST(P>0.0167), while its expression in breast adenosis tissues was lower than that in ILC and NST(P < 0.0167). In breast cancer the expression of EZH2 protein were not correlated with patient age, menopausal status, histological types, and pTNM stage. In contrast, its expression correlated with the tumor size, lymph node metastasis, molecular subtype and survival status(P<0.05). The positive rate of p53 protein in ILC, NST and breast adenosis tissues were 18.9%(18/95), 49.5%(198/400), 18.0%(9/50). There was no statistical significance of p53 between ILC and breast adenosis tissues(P>0.0167), while its expression in NST was higher than that in ILC and breast adenosis tissues(P<0.0167). Its expression had no related to patient age, menopausal status, tumor size, pTNM stage in breast cancer, but related to lymph node metastasis, histological types, molecular subtype and survival status(P<0.05). The expression of EZH2 protein was positively correlated with p53. The Kaplan-Meier survival analysis showed: the expression of EZH2 and p53 had correlated with disease-free and overall survival rates of breast cancer(P<0.05). Multivariate COX regression analysis showed that the expression of EZH2 was independent affecting factors to breast cancer patients. Conclusion: By detecting the relationship of EZH2 and p53 protein expression in breast adenosis tissues, ILC and NST, analyze their relationship between two expression and the clinical pathologic characteristics of breast cancer patients. To further explore the main effect of EZH2 and p53 protein in the evolution of breast cancer.
