Purpose:To investigate the affect of NOD2 to rapamycin (Rap) induced autophagy on proliferation and migration of tongue squamous carcinoma SCC-15 cells.Methods: â‘ NO2 expression vector (NOD2-pEZ-M29) was transfected into SCC-15 cells to increase NOD2 expression, NOD2-shRNA vector was transfected into SCC-15 cells to inhibition NOD2 expression.â‘¡ Rap to induce SCC-15 cells autophagy. Western blot was performed to examine the expression of NOD2,LC3 and Beclinl.MTT assay was used to evaluate cell proliferation. Wound healing was carried out to assess cell migration.Results:â‘ After Rap induce autophagy, the expression of NOD2,LC3 II and Beclinl in NOD2-pEZ-M29 group was significantly higher and markedly lower in NOD2-shRNA group.â‘¡Compared with control group, after Rap induce autophagy,the proliferation and migration ability were decreased in NOD2-pEZ-M29 group, and were increased in NOD2-shRNA group.Conclusion:NOD2 can up-regulate autophagy to suppress proliferation and migration in tongue squamous cell line.
