| Objects:To observe the effcts on general situation,ethology and hippocampus’ glucocorticoid receptor m RNA expression in rats with Mahuangfuzixixin Tang by copying the separation and chronic unpredictable mild stress depression rats model, investigate the association between the Mahuangfuzixixin Tang which represent the therapy of Wenyangxuanfei and GRm RNA expression in antidepressant treatment,provide the experimental data to understand the mechanism of Mahuangfuzixixin Tang in antidepressant treatment.Methods:1.Copy Model:The rats were fed with sepatation and chronic unpredictable mild stress(CUMS) to establish the animal models.There are seven stress methods in the CUMS program including fasting,water deprivation,clipping tail,forced icewater swimming,day and night reversed,damp bedding,thermal stimulation.We randomly gave a stress method everyday and make the stress methods appear discontinuously.We consistently give strss with separation by 35 days to copy the depression rats model.2.Grouping Disposal:We divided 50 SPF male Wistar rats into five groups including normal control group(NC),separation and chronic unpredictable mild stress one group(M1), separation and chronic unpredictable mild stress twogroup(M2),Mahuangfuzixixintang group(MT),mifepristone group(RU) by weight stratified randomly sampling.Every group contains ten rats.There are no stresses in the NC group and fed freely.We gave stresses to the M1,M2,MT and RU group for 35 days.M2 group was given stress for 70 days.After 35 days,the M1 and M2 group were given 3ml 0.9% Na Cl to fill the stomach daily.The MT group was given 3.0 g/kg to fill the stomach daily.The RU group was given 50 mg/kg to fill the stomach daily.We gave them to rats in 8:00-9:00 a.m. daily for 35 days.3.Observation:Make the general situation observation everyday and determine weight on the first day of a week.Do the rats’ behavior assessment in the 1st day,36 th day and 71 st day.Do the Morris water maze experiment from the 66 th day to the 71 st day. Detect the expression of rat hippocampal GRm RNA by realtime PCR in the end of experiment.Results:1.The general situation of rats was normal in the two weeks of adaptive breeding.And the result of rats’ behavior assessment has no statistical significance(P > 0.05).2.After stress for 35 days,all rats change significantly in the general situation including decreased activities,skin dryness and mood swings except the NC group.The results of M1,M2,MT,RU group in the open-field test and the sucrose preference test decreased significantly than the NC group(P < 0.05 or P < 0.01).It suggests that the model was copied successfully.3.In the end of experiment,the results of open-field test and the sucrose test have no statistical significance between the M2 group and the M1 group(P > 0.05).But the results have improved in the MT group and the RU group compared with the M1 group(P<0.05 或 P<0.01).4.In the Morris water maze test,the time and distance percent during platform quadrant of the M1 group and the M2 group have decreased(P < 0.05) and the escape latent period and the search distance of the M2 group have significantly prolonged(P<0.01 或 P<0.001) and the search distance of the RU group has prolonged than the NC group.5.In the realtime PCR detection,the expression quantity of the M1,M2 group reduced significantly than the NC group and it has varying degrees of increase in the MT,RU group than the M1 group.Conlusions:1.The Mahuangfuzixixin Tang has significantly antidepressant effct on the behavior of rats,and its mechanism may be related to up-regulate the GRm RNA in rat hippocampus.2.It is a good model combined the separation with chronic unpredictable mild stress(CUMS).So it can make a good foundation for the further research of the Mahuangfuzixixin Tang dealing with the depression.3.It can provide new ideas for clinical treatment of depression through the Mahuangfuzixixin Tang and the Wenyangxuanfei therapy. |
