Objective:To investigate the expression levels of fetuin-A and MGP in renal papillae tissues (Randall’s Plaque) in patients with calcium oxalate kidney stones.Methods:In our study,13 patients with calcium oxalate kidney stone in the experimental group and 12 individuals of renal tumor in the control were involved. The renal papillae tissues were obtained from patients in the experimental group and the control during operations. The expression of fetuin-A and MGP mRNAs in renal papillae tissue were evaluated by real time-polymerase chain reaction (RT-PCR). And the protein expression of fetuin-A and MGP in renal papillae tissue were localized and evaluated by immunohistochemistry.Results:1.The expression of Fetuin-A mRNAs in the experimental group was lower than that of control (P<0.05), while the expression of MGP mRNAs in the experimental group was higher than that of control (P<0.05).2.The immunohistochemistry showed that the protein expression of Fetuin-A in the experimental group was lower than that of control (P<0.05),and the protein expression of MGP in the experimental group was higher than that of control (P<0.05).3.in the thin segment of renal tubular and the colletcting duct, the expression level of Fetuin-A in the experimental group was lower than that of control (P<0.05), while the MGP was higher than that of control (P<0.05).Conclusions:1. Our data revealed that calcium oxalate kidney stone patient had a lower Fetuin-A expression level and a higher expression level of MGP than the control group in renal papillae tissue, a lower level of Fetuin-A and a higher expression level of MGP in renal papillae tissue may be a risk factor for calcium oxalate kidney stones. And the different mechanisms in regulating the formation of Randall’plaque of Fetuin-A and MGP may be responsible for the differences between the expression of MGP and Fetuin-A in renal papillae tissue.2. The decreased expression levels of Fetuin A in the experimental group in the thin segment of renal tubular and the collecting duct may be closely associated with the inflammatory response. And the increase expression level of MGP in the experimental group in the thin segment of renal tubular and the collecting duct may be associated with stimulation of renal papillary calcification and the crystals.
