Objective:To investigate the change of the expression of toll-like-receptor 4/nuclear factor-KB mRNA and protein and the effect of exercising on the expression of toll-like-receptor 4/nuclear factor-KB in the kidney of diabetic atherosclerosis Sprague-Dawley rat, the diabetic atherosclerosis rats model were established by using high-glucose and high-fat diet plus streptozotocin.Methods:(1) 50 male SD rats were randomly divided into the control group (CG, n=10) and the model groups (MG, n=40), subsequently the model group were randomly divided into 4 subgroups. After fed with high-glucose and high-fat diet for 4 weeks, the rats in model groups were injected STZ intraperitoneal injection. Continued the same diet for 6 weeks, the aorta tissues were observed to confirm whether the atherosclerosis model was established sucessfully. Six weeks of swimming exercise in the large physiological load of exercise group (LEG) and the moderate exercise group (MEG) and the small amount of exercise group (SEG) in a day respectively were 30minutes/ 21 minutes/12minutes.(2) Fasting blood glucose was measured by Roche glucose meter and related blood glucose test strips; the structure of aorta tissues which were made by HE staining were observed by using microscope; the TLR4/NF-ΚB mRNA and protein expression in kidney tissue were determined by RT-PCR and immune-histochemistry.Result:(1) Glucose:BEFORE EXCISE:1) the glucose in CG is significantly lower than other four groups, p<0.05; the glucose has no difference among the model control group (MCG), SEG, MEG and LEG, P>0.05. AFTER EXERCISE:1) the glucose in the control group is significantly lower than other four groups (p=0.000); 2) the glucose in MEG is significantly lower than MCG (P=0.003). BEFORE AND AFTER EXERCISE:1) the glucose in CG has no change (P=0.452); 2) the glucose measured in the 19th week is higher than 13th in MCG (P=0.001)、SEG (P=0.003)、MEG (P=0.007)、LEG (P=0.017).(2) Weight:1) AFTER 4 WEEKS HIGH-GLUCOSE AND HIGH-FAT DIET:the weights in LEG、MEG、SEG were significantly higher than CG,P=0.000; the weight has no difference among the MCG, SEG, MEG and LEG (P>0.05).2) AFTER EXERCISE:compare with CG, the weights of MCG、LEG、MEG、SEG were lower, P=0.000; the weight of MEG was higher than MCG,P=0.041.3) BEFORE AND AFTER EXERCISE:except the CG, the weights of the other four groups in the 19th week were lower than weights in the 19th week,P=0.000.(3)The aorta tissues which were made by HE staining were observed by using microscope in all groups were noticed that they had different levels of atherosclerosis except for the CG.(4)The TLR4 mRNA and protein expression:the TLR4 mRNA and protein expression of MG, SEG, MEG, LEG are higher significantly than CG (P<0.05) while they are lower significantly in SEG, MEG, LEG than MG (P<0.05).(5)The NF-ΚB mRNA and protein expression:the TLR4 mRNA and protein expression of MG, SEG, MEG, LEG are higher significantly than CG (P<0.05) while they are lower significantly in SEG, MEG, LEG than MG (P<0.05).Conclusions:High sugar and high fat diet can promote the progression of atherosclerosis in SD diabetic rats; the expression of TLR4 and NF-ΚB will increase in the kidney tissue of diabetes atherosclerosis rat. Exercise can reduce the expression of TLR4 and NF-ΚB in the kidney tissue of diabetes atherosclerosis rat; diabetes atherosclerosis is associated with TLR4 and NF-ΚB. Exercise has significant effect on the occurrence and progress in the in diabetic atherosclerosis.
