Objective To study the effects of vascular endothelial growth factor (VEGF), angiopoietin-1 (Ang-1) and angiopoietin-2 (Ang-2) on expression of neuregulin-1 (NRG-1) and secretion of NRG-1β in human cardiac microvascular endothelial cells (HCMEC)Methods ErbB2, ErbB3, and ErbB4 protein expressions were measured by Western bolt. Normal culture and hypoxic and deficient serum culture, 100ng/ml of VEGF, Ang-1 and Ang-2 cytokine were respectively added into the HCMEC, and detected expression and secretion of NRG-1 with Western blot and ELISA after 24 hours incubation.Results ErbB2, ErbB3 and ErbB4 were expressed in HCMECs. Under normal culture conditions, In VEGF and Ang-1 Treatment groups, the expression of NRG-1 and the secretion level of NRG-1 β were significantly increased compared with the control group, (P< 0.05), While these in Ang-2 treatment group were significantly decreased compared with the control group (P< 0.05). Under hypoxic and deficient serum culture conditions, In hypoxic and deficient serum group, the expression of NRG-1 and the secretion level of NRG-1β were significantly increased compared with the control group, In VEGF and Ang-1 Treatment groups, the expression of NRG-1 and the secretion level of NRG-1β were significantly increased, (P<0.05), whereas those in Ang-2 treatment group were significantly reduced compared with hypoxic and deficient serum group (P<0.05).Conclusion With VEGF and Ang-1 treatment, the NRG-1 expression and NRG-1β secretion in HCMEC are significantly increased, while with Ang-2 treatment, These are significantly decreased.
