ObjectiveOur aim was to research the effects of acupuncture on MAPK Signal Transduction Pathway of vascular endothelium on OLETF Rats, in order to further explore the potential mechanism of acupuncture on improving endothelial dysfunction.MethodsThe model rats were randomly divided into three groups:control group (LETO rats, n=9), model group (OLETF rats, n=9) and electro-acupuncture group(EA-OLETF rats, n=9). A group of the OLETF rats were treated with electro-acupuncture (EA-OLETF rats). The acupuncture was applied to bilateral Neiguan (PC6), Zusanli (ST36), Sanyinjiao (SP6), and Shenshu (BL23). Neiguan(PC6), Zusanli (ST36), and Sanyinjiao (SP6) were subcutaneously needled for 8 mm, and Shenshu (BL23) perpendicularly for 5 mm. Needles in Sanyinjiao (SP6) and Zusanli (ST36) were connected with electro-acupuncture apparatus, with sparse-dense wave (spares wave 2 Hz, dense wave 50 Hz,6 s, respectively) for 20 min, once a day, for 21 days. Throughout the experiment, using glucose oxidase method, ELISA, Western-blotting methods to detect fasting insulin (FINS), the fasting blood glucose (FBG), HOMA-IR, serum Opeptide (C-P)、NF-α、ET-1、IL-6、CRP、ICAM-1、VCA-1、PAI-1 and protein expression of MAPK in vascular endothelium of comparison group rats.ResultsFPG of model control group significantly increased compared with blank control group (p<0.01, p<0.01), FINS, C-p increased, compared with blank control group HOMA-IR than the control group has a rising trend. Prompt OLETF rats have insulin resistance. The FPG of acupuncture group decreased significantly in the model group (p<0.01), FINS, C-p compared with model group showed a trend of decrease, HOMA-IR showed a trend of decrease in the model group, the FPG was significantly reduced (p<0.01). Show that acupuncture improves OLETF rats with insulin resistance, and make it tend to be normal. Model group rats vascular endothelial MAPK-P38 lightning protein expression significantly higher than the control group (p<0.01); Acupuncture group rat vascular endothelial MAPK-P38 lightning protein expression decreased significantly in the model group (p<0.01), significantly lower than the control group (p<0.01).Serum level of TNF alpha model group was obviously higher than that of control group, and the model group and acupuncture group, level of TNF alpha OLETF rats were higher than dojdlivoe-leto wonders in rats (p<0.05), the level of TNF alpha was inersely associated with homa-ir were positively correlated, suggests that TNF alpha involved in T2DM and obesity-related IR.ConelusionAcupuncture intervention make insulin resistance model rats of TNF alpha reduce, inhibiting the activation of MAPK-P38 lightning signals at the same time, further reduce the production of TNF alpha, thus improve the activity of insulin receptor, increase insulin action, which may be of acupuncture to improve based on endothelial dysfunction is one of the mechanism of action of insulin resistance.
