Background Hepatocellular carcinoma is one of the most commonly diagnosed malignancy worldwide and is the main cause of cancer-related mortality. Every year,300000 people suffer from hepatocellular carcinoma in china and accounting for 60% of the world in patients with HCC. With the development of modern medicine, we established the principles of comprehensive treatment of HCC and given priority to with surgical treatment, however, due to the high morbidity, high recurrence rate, early mortality for HCC, Mortality rates of HCC are remain high, so we need to find some sensitive indicators, and use to diagnose and assess the prognosis of the patients of HCC earlier and more.Recently, much attention has been focused on the role of transmembrane protease serines (TMPRSSs), also called to type II transmembrane serine proteases (TTSPs), which are members of the family of cell surface-associated proteases. Increasing evidence demonstrates that aberrant expression of TTSPs is a hallmark of several cancers and recent studies have defined molecular mechanisms underlying TTSP-promoted carcinogenesis. TMPRSS4 is a novel TTSPs found at the cell surface that is highly expressed in pancreatic, colon, thyroid and gastric cancer tissues. TMPRSS4 was suggested as a potential diagnostic and prognostic marker for those cancer types. However, the biological functions of TMPRSS4 have not yet been elucidated in hepatocellular carcinoma (HCC). In this study, we can research the long-term survival relations with the level of TMPRSS4 expression after curative resection in patients of hepatocellular carcinomaAims The Long-term survival relations with the level of TMPRSS4 expression after curative resection in patients of hepatocellular carcinomaMethodology We analyze the commercially available tissue microarray (TMA) provided by the manufacturer (Shanghai Outdo Biotech Company) containing 398 pairs of HCC was used. The TMA sections were used for immunochemistry staining. According to result of immunochemistry staining, we could evaluated TMPRSS4 expression based on these results and divided into two groups:TMPRSS4 express positive group 128 and express negative group of 270 people. Compared the two groups of patients and using statistical methods to analyze whether the differences between two groups have statistical significance. Patients’ demographic data, residual tumor were similar in both groups. Overall morbidity was similar in the two groups.Result TMPRSS4 expressed in hepatocellular carcinoma cells is higher than adjacent nontumor cells (P<0.05). Expression of TMPRSS4 shows a positive correlation with tumor size (>5cm,38.5% vs ≤5cm,27.5%, p<0.05), TNM staging (â…¢ staging,50.0% vs â… -â…¡ staging,29.8%, p<0.05) and vascular invasion (invasion, 40.6% vs non-invasion,29.3%, p<0.05). The 1-,3-, and 5-year OS rates after curative resection for TMPRSS4 positive patients were 82.2%,50.0% and 34.9%, significantly worse than those of TMPRSS4 negative patients (93.3%,75.6% and 60.9%), p<0.05 was considered statistically significant.Conclusion TMPRSS4 expressed in hepatocellular carcinoma tissue is higher than adjacent nontumor tissue. Expression of TMPRSS4 shows a positive correlation with tumor size, TNM staging and vascular invasion. Expression of TMPRSS4 is predictive of poor prognosis in HCC patients after curative resection. |
