As we know, breast feeding is more and more important and is accepted By us all day by day. Breast milk can provide the infants with all kinds of nutrient element. Adequate secretion of breast milk becomes a necessary condition for an infant’s health and growth. Nowadays, the incidence rate of postpartum hypogalactia becomes higher and higher.There are many methods of treating postpartum hypogalactia, for example, western and traditional Chinese medicine, Chinese diet therapy, acupuncture andmoxibustion, manipulation, physiotherapy and so on.By comparison, we can find that, Chinese diet therapy, diet nursing, neither has the problem of serious side effect of western medicine, nor has the problem of highfee.Therefore, traditional Chinese diet therapy’s development prospect is very good. As a kind of traditional Chinese diet therapy method of diet nursing, chicken rice wine’s applying history in helping treating postpartum hypogalactia is quite long. To a pity, its science basis isn’t found yet. And chicken rice wine’s isn’t reported by formal periodicals up to now. This study is about giving chicken rice wine to the mummy rats which are postpartum hypogalactia by gavage, then we observe whether chicken rice wine can improve the secretion of breast milk. Furthermore, this study has a primary discussion about the mechanism treatment effect of chicken rice wine on postpartum hypogalactia on rats. Then we can provide a convictive basis on the clinical application and promotion of chicken rice wine.Objective:The aim of this study is to discuss the mechanism of treatment effect of chicken rice wine on postpartum hypogalactia rats, and to evaluate that whether chicken rice wine has improvement effect on the secretion of Prolactin and β-casein kinase of postpartum hypogalactia. At the same time, we eagerly want to find convictive scientific basis on how chicken rice wine treat postpartum hypogalactia, then we can provide important clinical reference for chicken rice wine’s promotion.Methods:Select 40 6~9 months SPF pregnancy BALB/c rats and 14 6~9 months SPF female unpregnancy BALB/c rats,total 54. Then divide groups, first, we divide 14 unpregnancy rats into two groups randomly. Group A are given normal diet. Group B are given chicken rice wine by gavage on the basis of normal diet. Second, about the 40 pregnancy rats their time to get pregnancy is very close. After they give birth to their children, then we select 28 mummy rats which are delivering in the same 24 hours to our study. At the same time, every mummy rats raise six its own children(baby rats). And then they are divided into four groups randomly. Group C and Group D are normal postpartum groups. Group C are given normal diet. Group D add gavaging chicken rice wine on the basis of normal diet.The other two groups (Group E and Group F) sre given 0.05mg/mL bromocriptine to become medels of pastpartum hypogalactia mummy. The dose is lOmL/kg every day, totally for seven days. Group’s diet is the same as Group C. Group’s diet is the same as Group D. And for Group B、 D、F, the dose of gavaging chicken rice wine change with weight every day. it’s also lOmL/kg, one time a day, and the tatal gavage time is one week. Afer one’s gavage, we select blood serum and breast tissue the next day (the ninth day after delivering). Afer that, we detect the rats’concentration of prolactin in the serum through the method of Enzyme Linkeed Immunnosorbent Assay. Then, we observe the histological changes of mammary glands of the rats by HE stain. W e also test the expression of Prolaetin Receptor and β-casein kinase, then we analyze the expression levelof Prolactin Receptor and p-casein kinase through using IPP 6, a kind of image analysis sofeware.Results:Through comparing the concentration of prolactin in the rats’serum, we find that, between Group E and Group F, the concentration of prolactin those which are given chicken rice wine is all higher than the normal diet groups (P<0.05).But, among the groups of unpregnancy rats and the normal pastpartum rats, the concentration of prolactin of those which are given normal diet is higher than the groups which are given normal diet and chicken rice wine (P <0.05).Through observing the mammary glands of these rats under light microscope,we can find that, the mammary glands the rats which are given chicken rice wine differ from the mammary glands of the rats that are given normal diet.There are some differences, the first one is that the number of mammary glands bubbles is more than the narmal diet groups.The second difference is the lumen diameter of chicken rice wine groups is abviously bigger than the normal diet groups. Then the immunohischemical stain shos that the expression level of prolactin receptor in rats’ mammary glands is higher than the normal diet groups. And the β-casein kinase’s result is the same as prolactin receptor. The Semi-quantitative image analysis tells us that the positive area and the integral optical density of prolactin receptor and β-casein kinase in the rats’ mammary glands of chicken rice wine groups are both higher than the normal diet groups (P<0.05)Conclusion:Chicken rice wine can effectively promote maternal lactation of the postpartum hypogalactia rats, the mechanism may be chicken rice winecan stimulate the secretion of prolactin, then increase the expression of prolactin receptor, this can help the epithelial cell proliferation of mammary glands promote and the breast develops faster, too. Therefore the secretion of breast milk increases. In summary, chicken rice wine can promote breast milk’s secretion,we find a scientific basis for that through this study. We can conclude that chicken rice wine is a good method of treating postpartum hypogalactia.
