Objective:1.To investigate the expressions of Twist in breast carcinoma and breast fibroadenoma by Immunohistochemistry and explore the relationship between Twist and Clinicopathological Characteristics of breast carcinoma patients.2.Western blot was used to detect the expression of Twist protein in breast carcinoma and breast fibroadenoma.3.To analyze the predictive factors of axillary lymph node metastasis in breast carcinoma, provide experimental and theoretical basis for correct clinical staging as well as comprehensive treatment.Methods:1.We collected formalin fixed paraffin-embedded tissues of 82 cases diagnosed with invasive breast carcinoma by the Pathology Department of the Affiliated Hospital of Luzhou Medical College in January 2014 to October 2014. Collected and combined the medical history of all the patients who had their first episode as well as no history of treatment(including chemotherapy, radiotherapy or endocrinotherapy). In addition, 22 cases of breast fibroadenoma were collected.2.Immunohistochemical En Vision stains of Twist were performed in breast carcinoma and breast fibroadenoma respectively. SPSS 19.0 was used to analysis all the dates.3.Re-checked the paraffin section and reviewed all the breast carcinoma diagnosis, then re-evaluated the histologic grade and nuclear grade.4.Consulted medical records and pathological reports. Clinicopathologic datas such as age, tumor size, menstrual status, ER, PR, Her-2, Ki-67, TNM staging and axillary lymph node status were collected. Used SPSS 19.0 to analyze the correlation between levels of Twist and the above indicators and predicted the factors of axillary lymph node metastasis.5.Fresh breast tissue were collected from the Affiliated Hospital of Luzhou Medical College in December 2014 to March 2015. Breast carcinoma tissue and breast fibroadenoma tissue were included.The expression of Twist proteins were detected by Western Blot.Results:1.Immunohistochemical results show: the positive expression of Twist protein was detected in the nucleus of cells in breast tumor. In the 82 cases of breast carcinoma, the positive expression were 62 cases and the positive rate was 75.6%. In the 22 cases of breast fibroadenoma, the positive expression were 7 cases and the positive rate was 31.8%. There was a statistically significant differencebetween them(P<0.05).2.The expression level of Twist in breast carcinoma was relevant to the tumor size(P=0.019), histological grade(P<0.001), nuclear grade(P<0.001), TNM stage(P=0.006), ER(P=0.022), Ki-67(P=0.010) and axillary lymph node metastasis(P=0.038), while there was no obvious correlation among age( P=0.700), PR(P=0.177), Her-2(P=0.739) and menstrual statusand(P=0.890).3.Western Blot results showed that the expression of Twist protein in breast carcinoma were significantly higher than breast fibroadenoma, and these results were consistent with the IHC result.4.The single factor analysis on axillary lymph node metastasis indicated that there was a significant correlation with tumor size(P=0.043), histological grade(P=0.002), nuclear grade(P=0.041) and Twist(P=0.038)while there was no correlation between age(P=0.191), menstrual status(P=0.221), ER(P=0.976), PR(P=0.728), Her-2(P=0.758)and Ki-67(P=0.795).Conclusions:1.By IHC and Western Blot, the results showed that Twist was expressed highly in breast carcinoma while expression in breast fibroadenoma was low.2.Comparing Twist with clinicopathological factors, we discovered a significant correlation among tumor size, histological grade, nuclear grade, ER, axillary lymph node metastasis, Ki-67 and TNM stage,the result confirmed that Twist could increase the capacity of invasion and metastasis in tumor. Testing the expression levels of Twist might provide theoretical basis for clinical diagnosis and staging, may also provides reference for prognosis of breast carcinoma patients.3.Twist, Tumor size, histological grade and nuclear grade can be regarded as the predictive factors of axillary lymph node metastasis in breast carcinoma patients. |
