Objective: Effect of curcumin on proliferation, cell cycles, apoptosis, of human esophageal carcinoma cell Eca-109 and mutuality downstream gene of Wnt/β-catenin signaling pathway were studied. To produce scientific evidencefor well developing curcumin.Methods:1 The suppressive effect of curcumin on the proliferation of Eca-109 cell among different concentration and different time were analyzied by MST method.2 After treatment with curcumin under different concentration(10μmol/L, 20μmol/L,40μmol/L) for 24 and 48 and 72 h, the cell cycles and apoptosis rate of Eca-109 cell were detected by flow cytometry(FCM), and the expression changes of cyclin B1 was investigated by Q-PCR and western-blot.3 After the intervention of curcumin for 48 h, gene expression and protein content of β-catenin and GSK-3? and c-myc of Wnt/β-catenin signaling pathway were detected by Q-PCR and western-blot.Results:1 MTS assayed showed, after treatment with curcumin under different concentration(10μmol/L, 20μmol/L, 40μmol/L) for 24 and 48 and 72 h, inhibition rate of Eca-109 cells proliferation is on the rise.The more the concentration of curcumin, proliferation inhibition rate was more obvious.And a dose-dependent relationship can been seen between them(P<0.05).2 The result of flow cytometry analysis indicated that, after treatment with curcumin for 48 h, the cell cycles and apoptosis rate of Eca-109 cells were changed,G2/M phase were increased(P<0.01) and G0/G1 phase were reduced(P<0.01).Apoptosis rate increased significantly(P<0.01).With increasi ng concentration of curcumin,the difference was more obvious.The result of Q-PCR and Western blot indicated that gene expression and protein content of Cyclin B1 were reduced(P<0.05).3 The result of Q-PCR and Western blot indicated that, compared with the control group,gene expression and protein content of β-catenin and c-myc were reduced,the difference was more obvious(P<0.01);gene expression and protein content of GSK-3? were increased,the difference was more obvious(P<0.01).Conclusion:Curcumin can cause G2/M phase arrest in Eca-109 cells, lead to restrain proliferation, and induce apoptosis in human esophageal carcinoma cell.Curcumin may be through up-regulation of expression of GSK-3?, reduce expression of ?-catenin,block the signaling pathway,inhibite expression of target gene c-myc, lead to restrain proliferation, and induce apoptosis in human esophageal carcinoma cell.
