Objective: To detect the chemoresistant related proteins in the serum among patients with epithelial ovarian cancer, expecting to find the protein biomarkers relating to the chemoresistance of epithelial ovarian cancer.Methods: Collecting the patients’ plasma samples who were diagnosed and treated with surgery and chemotherapy in the fourth hospital of Hebei Medical University from September 2013 to September 2014. We screened the differentially expressed proteins by using two dimensional gel electrophoresis(2–DE). And then, comparing the protein points between the chemo-sensitive group and the chemoresistant group, got the different protein points which were further analysised and identified by using matrix-assisted laser desorption/ionization time of flight mass spectrometry( MALDI-TOF–MS).Results: 1 The chemotherapy resistance was related to staging of ovarian cancer by chi-square(Correlation coefficient =0.77), but it has no obvious correlation with the types of pathology(Serous, Endometrioid and clear cell carcinomas), pathological grading and age. 2 34 differentially expressed points were identified between the chemo-sensitive group and the chemoresistant group. Compared with the chemo-sensitive group, 21 protein points were up-regulated and 13 protein points were down-regulated in the chemoresistant group, but only 14 possible differentially expressed points were found by using the Mass spectrometry and Mascot searching, which were Complement C4-A, Transthyretin, Immunoglobulin J chain, Clusterin, Ig kappa chain C region, Alpha-1-antitrypsin, Complement C3, Haptoglobin, Beta-2-glycoprotein, Ig gamma-2 chain C region, Ig gamma-1 chain C region, Carbonic anhydrase 1, Complement factor I light chain, Apolipoprotein E. Compared with the chemosensitive group, Complement C4-A, Immunoglobulin J chain, Clusterin, Alpha-1-antitrypsin and Carbonic anhydrase 1 were upregulated, and Transthyretin, Haptoglobin, Beta-2-glycoprotein, Ig gamma-2 chain C region, Ig gamma-1 chain C region, Complement factor I light chain, and Apolipoprotein E were down-regulated in the chemoresitant group.Conclusion: The upregulated expressions of Transthyretin, Apolipoprotein E and Haptoglobin proteins and the down-regulated expressions of Clusterin, Carbonic anhydrase 1, Alpha-1-antitrypsin were significantly differentiated between the chemo-sensitive group and the chemoresistant group, which maybe pretentially used for predicting the sensitivity of chemotresistance in epithelial ovarian cancer patients.
