Objective: To investigate the effects of crystal solution, colloidal solution and crystal glue solution in restrictive resuscitation on interleukin(IL)-4, IL-10 and T lymphocytes in rats of hemorrhagic shock.Methods: According to random number table, 24 healthy SD rats were divided into four groups: one normal control group and three experimental groups, the latter contains sodium Lactated Ringer’s group(crystal group), Hydroxyethyl Starch group(colloidal group), sodium Lactated Ringer’s and Hydroxyethyl Starch group(crystal glue group). The hemorrhagic shock models were established in the three experimental groups by bleeding through the femoral artery. Draw blood of rats in each experimental group for five times: before shock(T0), after 30 minutes of keeping MAP in 40 mm Hg(T1), after 30 minutes of keeping MAP in 60 mm Hg(1 mm Hg=0.133 k Pa)(T2), after 60 minutes of keeping MAP in 80 mm Hg(T3), and at 24 h after successful resuscitation(T4). The rats of normal control group were only drew blood at the same time when T0 and T4 came. ELISA method was used to measure IL-4 and IL-10 content in plasma. Flow cytometry method was used to measure T lymphocyte content.Results:1. Among all groups, there were no significant differences in the age, weight and MAP before experiment(P>0.05).2. The survival rate of the crystal group was higher than that of the crystal glue group or colloidal group, the difference had statistical significance(P<0.05).3. Before shock(at T0), the contents of IL-4 and IL-10 in plasma of each group had no statistical significance(P>0.05).When the shock began(at T1 and T2), the contents of IL-4 and IL-10 in plasma of each experimental group rats had no statistical significance(P>0.05). During the restrictive resuscitation, the contents of IL-4 and IL-10 in crystal group and colloidal group were decreasing. The content of IL-4 and IL-10 in crystal glue group were higher than that of the crystal group or colloidal group at T3 and T4, the difference had statistical significance(P<0.05).4. Before shock(at T0), the ratio of CD4+ T cells to CD8+ T cells in each group had no statistical significance(P>0.05).When the shock began(at T1 and T2), the ratio of CD4+ T cells to CD8+ T cells in plasma of each experimental group had no statistical significance(P>0.05). During the restrictive resuscitation, the ratio of CD4+ T cells to CD8+ T cells in crystal group and colloidal group were decreasing. the ratio of CD4+ T cells to CD8+ T cells in crystal glue group were higher than that of the crystal group or colloidal group at T3 and T4, the difference had statistical significance(P<0.05).Conclusion:1. In the process of restrictive resuscitation,it may decrease survival rate when we treat the rats only by crystal solution.2. During the restrictive resuscitation, crystal glue solution can better accelerate the release of anti-inflammatory mediators and regulate the immune function.3. Crystal glue solution may be a relatively better restrictive resuscitation liquid. |
