Objective: Study the formation of bilateral memory caused by unilateral learning in mice, and observes the changes of the intrinsic properties and synaptic transmission efficiency including the excitatory and inhibitory neurons in the barrel cortex.Methods: 1.Building: choose the mouse that was born 10 days of C57BL- 6- GFP,while giving a 5 Hz stimulation of the right beard along with another stimulation of butyl acetate smell, two hours at a time, five times a day, continuous training for 10 days.2. Behavior experiment: the experiment was divided into two groups, respectively for the control group and the associative learning and memory group. Behavioral experiments including the T maze experiment and the beard frequency. The T maze experiment: Having the T maze experiment with the mice of the associative learning and memory group and the control group respectively, namely the mice immediately entering the T maze after giving the 5 Hz stimulation of the right beard, recording the side direction of avoiding butyl acetate in mice. Beard frequency experiment, during the period of building, after giving the sense stimulation of smell immediately shooting video, recording the bilateral beard frequency of the two groups respectively by using the video studio software.3. Electrophysiological experiments: Through testing to determine whether a model is established successful, then letting the successes building of the associative learning and memory group and the control group mice have the electrophysiology experiment.Following the detailed process of the electrophysiology experiment: anesthetized the mice with isoflurane, rapidly cut the head removed brain,then made the coronary slice by the pathological slicer machine(400 microns);putting the slices into the oxygenation of artificial cerebrospinal fluid(ASCF) which the temperature is 25 ° C;45 to 60 minutes later, transferring them to the perfusion tank, perfusion of brain slice in 30 ° C; Application of optical microscope and fluorescence microscope localization the glutamate neurons and GABA neurons in the barrel area of bilateral; Using Axon-clamp–200B amplifier and D/A converter, recording the current and the voltage through the whole cell clamp mode, studying the changes of the inherent characteristics and the synaptic transmission efficiency of the glutamate neurons and the GABA neurons in the barrel cortex of bilateral, then analysis the statistics by using the Clamp 10.1.Results: 1. the results of behavioral experiments A) The results of T maze experiment: the probability of avoiding the butyl acetate of the associative learning and memory group is higher than the control group mice;B) Beard frequency experiment results: both side of the associative learning and memory group mice the frequency of beard were higher than the control group mice,and the frequency of the right side of the beard is higher than the left side of the beard.2. The results of electrophysiology experiments A) The result of Current clamp1) Pyramidal neurons: compared with the control group, the energy barrier of the neuron in the barrel cortex corresponding to both side whiskers of the associative learning and memory group was reduced, the distance of the action potential was shortened, the absolute refractory period was shortened, the issue number of the action potential per unit time was increased; And on the right side of beard corresponding barrel cortex compared with the contra-lateral beard barrel cortex, the energy barrier was reduced and the distance of action potential was shortened, absolute refractory period was shortened, the issue number of action potential per unit time was increased.2) GABA neurons: compared with the control group, the energy barrier of the neuron in the barrel cortex corresponding to both side whiskers of the associative learning and memory group was raised, the distance of the action potential was increased, the absolute refractory period was extended, the issue number of the action potential per unit time was reduced; And on the right side of beard corresponding barrel cortex compared with the contra-lateral beard barrel cortex, the energy barrier was higher,and the distance of action potential was increased, the absolute refractory period was extended, the issue number of action potential per unit time was reduced.B) The result of voltage clamp1) Pyramidal neurons: compared with the control group, the s EPSC interval of events on the both side of the barrel cortex of the associative learning and memory group was shortened, and the amplitude was increased; and on the right side of beard corresponding barrel cortex compared with the contra-lateral beard barrel cortex,s EPSC interval of events was shortened, and the amplitude was increased. Compared with the control group, the s IPSC interval of events on the both side of the barrel cortex of the associative learning and memory group was increased, the amplitude was decreased; and on the right side of beard corresponding barrel area compared with the contra-lateral beard barrel area, s IPSC interval of events was increases, the amplitude was decreased.2) GABA neurons: compared with the control group, the s EPSC interval of events on the both side of the barrel cortex of the associative learning and memory group was increased, the amplitude was decreased; And on the right side of beard corresponding barrel area compared with the contra-lateral beard barrel cortex, s EPSC interval of events was increased, the amplitude was decreased.Conclusion: Memory can be stored in bilateral after unilateral study; and this kind of storage methods was asymmetric storage.
