Brucine is the main component of Strychnos nux-vomica Linn, it has not only pharmacological effects, but also toxicity. Thus it is very important to strictly control the content of Brucine in various samples.The study is focused on the determination method of Brucine in preparations and serum by HPLC. The solide phase extraction method was used to separate and purify the samples. The Brucine is slightly soluble in water. In order to improve the solubility, the inclusion compounds of Brucine with-β-cyclodextrin and HP-β-cyclodextrin were prepared. The solubility of the inclusion compounds was improved greatly.This paper includes five parts as follow:In chapter one, the application prospect, the separation and inspection methods of Brucine was reviewed. And the research of inclusion compounds was also reviewed.In chapter two, the HPLC method to determine the Brucine was established through screening of wavelength, mobile phase, temperature and other conditions. The method was validated and has good separation and precision, and the repeatability was conformed to meet the requirements.In chapter three, by comparison of the result of brusine separation by different solid phase extraction column, the appropriate extraction column, leacheate and eluent were chosen. The SPE-HPLC was applied to detect Brucine in ‘Fengshi Maqian’ tablet and the method has been proven to be handled simply, accurately and fast.In chapter four, the preparation of molecularly imprinting column was made by the bulk polymerization process using Brucine as the template molecule, Methacrylic acid as the functional monomer, EGDMA as the crosslinking agent, 2,2-Azobis as initiator, and chloroform as porogen. And then the results of extraction of the Brucine by molecularly imprinting extraction column and PXC column is compared, the experiment results show that the PXC solid phase column was suitable for the extraction of Brucine from biological samples.In chapter five, the saturated water solution method and freeze drying method were used for preparation of Brucine-β-cyclodextrin Inclusion compound and Brucine-HP-β-cyclodextrin Inclusion compound, separately. And the Brucine Inclusion compounds was characterized of by Ultraviolet spectrophotometry, Infrared spectroscopy and Thin-layer chromatography. The Brucine Inclusion compound has improved the water solubility of Brucine, significantly.
