Objectives The purpose of this study was to investigate the inhibitory effect of betaine onendotoxin-induced uveitis and mechanism in rats.Method Sixty Sprague-Dawley rats were used, the rats were randomized into five groups oftwelve rats each. The three treatment groups were administered betaine (100,200and400mg/kg)for30days, the control group and the LPS group were administered the same volume of salinefor30days. EIU was induced by footpad injections LPS.24h after the LPS injection, the ratswere sacrificed. Ocular tissues were collected for a histopathological evaluation. The level ofiNOS and eNOS in eye tissue, level of serum nitric oxide and ADMA,activities of serumsuperoxide dismutase(SOD) and glutathione peroxidase(GSH-PX),and content ofmalondialdehyde(MDA), and alanine aminotransferase (ALT), aspartate aminotransferase theenzymatic (AST), creatinine (CERA), triglyceride (TG)total cholesterol (CHOL), high-densitylipoprotein cholesterol (HDL), low density lipoprotein (LDL) cholesterol concentration weredetermined by using Western Blot,ELISA and biochemical methods.Results According to the pathological study,bataine(100,200,400mg/kg) could notably reducethe inflammatory cell infiltration around corpus ciliare matrix of uveitis rats, also inhibit theexpression of iNOS and increass the expression of eNOS in oculartissues.Betaine(100,200,400mg/kg) significantly reduce the concentration of protein and nitric oxide,and increased ADMA in serum.In addition,betaine(100,200,400mg/kg) also decrease theactivities of serum SOD,GSH-PX,and the content of lipid peroxide MDA, and increase the level ofALT, AST, TG, CHOL, HDL, LDL in serum,and reduce the level of CERA.Conclusion Betaine has the inhibitory effect on LPS-induced uveitis in rats. Its mechanism isrelated to the regulation of NO/ADMA pathway and the improvement of oxidative stress, alsoassociated with Lowering blood lipids, protect the liver and kidney.
