Objective To clarify GABA neurons expression in the GAD67-GFP mousehippocampus of schizophrenia and detect GABA neurons and granule cell layer of the dentategyrus granule cells of newborn coexistence model for the study of the pathogenesis of SP toprovide new experimental and theoretical basis.Methods Application of NMDA receptor antagonist MK-801, was prepared animalmodel of schizophrenia. Two groups of animals were carried out BrdU labeling,decapitated deal:(1) Immunofluorescence staining:observe the expression of hippocampalGABA neurons and the relationship with newborn granule cells labeled with BrdU;(2) Nisslstaining. observe changes in hippocampal dentate gyrus granule cell layer neurons (3) Byflow cytometry: to observe the proliferation of hippocampal neurons.(4) Western-blottechnique: to detect changes of hippocampal GABA neurons protein levels.(5) RT-PCRtechnology: to detect changes of the hippocampus GAD67mRNA expression levels.Results (1) GABA neurons in the hippocampus:①Immunofluorescence stainingresults: the experimental group compared with the control group, the dentate gyrus GABAneurons showed no change in the number of positive cells (P>0.05); can not find coexistencebetween newborn granule cell neurons and GABA neurons.②Western-blot results: theexperimental group and the control group, GABAa protein and GFP protein have no significant change (P>0.05);③RT-PCR results: a significant decrease in the expression ofGAD67mRNA in hippocampus of mice in the experimental group than the control group (P<0.05).(2) Changes in the dentate gyrus neurogenesis:①Nissl staining results: theexperimental group and control group dentate gyrus line optical density analysis found nosignificant change (P>0.05), normal mice hippocampal dentate Nigeria ’s body back area neat,did not find its stained disorders, and abnormal increase; in the experimental group, dentategyrus Nissl bodies disorganized, broken, in the dentate gyrus granule cells stained underlyingseen a significant Nissl bodies accumulation zone.②Immunofluorescence staining results:the experimental group compared with the control group, the number of newborn granule celllayer of the dentate gyrus of the hippocampal neurons was significantly decreased (P <0.05)③flow cytometry results: the experimental group than in the control mice hippocampus ofmice cell proliferation decreased significantly (P>0.05);Conclusion (1) After the GAD67-GFP mice schizophrenia, hippocampal dentate gyrusneurons GABA-positive cell number did not change significantly.(2) After the GAD67-GFPmice schizophrenia, which can cause nerve cells in the dentate gyrus reduced proliferation,expression and arrangement of dentate gyrus Nissl bodies.(3) After the GAD67-GFP miceschizophrenia, at the protein level, the expression of GFP protein and GABAa protein in thehippocampus have no significant changes.(4) After the GAD67-GFP mice schizophrenia, atthe mRNA level, reduced GAD67mRNA expression in the hippocampus.
