Wolfberry Polygonatum prescription oral liquid was prepared, then stability studyand microbial limit was Investigated.3.To determine the resistance to hypoxia of mice by observing the survival time of themtook in oral liquid. To investigate the effect, adjusting the immune function of organism, ofWolfberry Polygonatum prescription oral liquid by determine the blood of mice carbonclearance rate and the weight of immune organs. Using acute high lipid model to determinethe content of TG, TC, HDL-C and LDL-C, so as to explore the antiatheroscloresis effect ofChinese wolfberry oral liquid.Result1. The yield of Wolfberry polysaccharide by reflux extraction is7.66%and the content of it is63.2%; that of Polygonatum polysaccharide are22.0%and70.0%. The yield ofWolfberry polysaccharide by ultrasonic extraction is3.41%and content of it is43.0%; that ofPolygonatum polysaccharide are14.53%and81.9%. The yield of polysaccharide ofwolfberry Polygonatum prescription by reflux extraction17.55%and the content of it is62.8%;the yield of that by ultrasonic extraction are12.92%and51.0%. The content ofpolysaccharide of wolfberry extracted by reflux and Polygonatum extracted by ultrasonic is70.0%; that of wolfberry extracted by ultrasonic and Polygonatum extracted by reflux is48.5%.2.The optimal preparation method is that: Antiseptic0.03%, Stabilization0.1%, pH7.Oral liquid is stable in temperature and light conditions. In accelerated test, the content of oralliquid decreased slightly after6months. There are no bacteria, fungi and yeast in the oralliquid.3.Compared with the control group, the group of high and medium dose of oral liquidcan prolong the survival time of mice in hypoxia condition, and there were significantdifferences (P<0.05); Compared with the control group, that of low dose improve bloodclearance rate, and there were significant differences (P<0.05); Compared with the controlgroup, that of high dose can increase the weight of immune organs, and there were significantdifferences (P<0.05); Compared with model group, that of high,medium and low dose all canreduce the content of TG,TC and LDL-f, increase the content of HDL-C (P<0.05).Conclusion1. The optimal extraction method of polysaccharide in wolfberry Polygonatumprescription is that wolfberry was extracted by reflux and Polygonatum was extracted byultrasonic, then the polysaccharides of both were merged. Reflux extraction technology ofWolfberry: the ratio of material to liquid is1:10, extracted twice and3.5h per time, thenprecipitated with80%alcohol. Ultrasonic extraction technology of Polygonatum: the ratio ofmaterial to liquid is1:20, extracted twice and20min per time, then precipitated with80% alcohol.2.Antiseptic content of wolfberry Polygonatum prescription oral liquid is0.03%.Stabilization content of that is0.1%. pH is7. The oral liquid is clear and brown, sweet, andthe antiseptic effect is well. Long term storage is convenient.3.Wolfberry Polygonatum prescription oral liquid has the effect of anti hypoxia,theregulation of immune function and antiatheroscloresis effect. |
