Relationship Between The GDF-9/BMP-15/IGF-Ⅰ Levels In Follicular Fluid And Patients With Polycystic Ovary Syndrome

Objective To investigate the levels of growth differentiation factor-9(GDF-9)、bonemorphogenetic protein-15(BMP-15)、insulin like growth factorⅠ(IGF-I)in follicular fluidin patients with polycystic ovary syndrome(PCOS)during ovulatiory cycle, To determinewhether there are differences about the levels of GDF-9、BMP-15、IGF-I in follicular fluidbetween PCOS patients and patients with normal ovulation. From molecular level to explorethe correlation between the three cytokines, GDF-9、 BMP-15、 IGF-I and the pathogenesisof PCOS and pregnancy outcome of in vitro fertilization and embryo transfer(IVF-ET).Methods The case-control study included60PCOS patients selected randomly and60PCOS-free controls, Western Blotting were conducted to detect the expression of GDF-9andBMP-15protein in follicular liquid which collected on the day of retrieval,Enzyme-linkedImmunosorbent Assay(ELISA) were used to determine the concentration ofIGF-I.Descriptive statistical analysis was performed by using SPSS13.0software, using χ2test for categorical variables, t-test for continuous variables and Pearson test for indexscorrelation.Results1.The concentration of serum LH, T, LH/FSH ratio and numbers of retrievedoocytes,high quality embryo number in PCOS groups were significant higher than controlgroup’s(p<0.05).2.The expression of GDF-9, BMP-15levels were significantly lower than control group (p<0.05).3.The expression of IGF-I, E2,T levels were significantly higher thancontrol group(p<0.05).4.The IGF-I level was negatively correlated with E2and T in bothgroups(p<0.05).5.The expression of GDF-9, BMP-15level in pregnancy patients werehigher than none pregnancy ones in both groups(p<0.05); In control group, the expressionof IGF-I levels in pregnancy patients were lower than none pregnancy ones(p<0.05).6.InPCOS group, the fertilization rate and high quality embryo rate of pregnancy patients issignificantly higher than none pregnancy ones (p<0.05).In the control group, high qualityembryo rate and cleavage rate of pregnancy patients is significantly higher than notpregnancy patients(p<0.05).Conclusions1.GDF-9,BMP-15and IGF-I are important factors in regulation ofovaries local autocrine and paracrine, they may involve in physiological and pathologicalprocess which led to disorder of follicular development.2.The high expression of GDF-9andBMP-15may be in favor of oocyte quality, embryo developmental potential and pregnancyoutcome.3.Too much IGF-I in follicular fluid may negatively effect pregnancy outcomes.4.IGF-I may play an certain role in the pathogenesis of PCOS by promoting E2and T synthesis.
