Objective:Apply particle gun eukaryotic expression plasmid containing the BMP-2gene local gene transfer in therapy of chronic bone defect. Methods Elect72healthy three months New Zealand white rabbits, set up old model of rabbit bone defect in middle of radial group rabbits according to the the defect length:(1)24rabbits in1.5cm group,(2)24rabbits in2.0cm group,(3)24rabbits in2.5cm group. Each group include two subgroups according to the intervention:12rabbits in treatment group (BMP-2gene transfection group) and12rabbits in control group. Three weeks later, through the application of particle gun for BMP-2bcal transfection after bone cutting, the control group with natural healing after bone cutting. X-ray examinations are taken at1week,3weeks,8weeks and9weeks after transfection, and the tissue between of the bone defect are taken to doing BMP-2Western blot detection at1week,3weeks,8weeks and9weeks; and radius specimens are taken for gross observation at1week,3weeks,8weeks and9weeks, evaluation of the healing. Results:(1) Gross specimen observation:bone callus formation of treatment group is generally more than the control group.(2)Lane--Sandhu X-ray score:there is statistically significant atl,3,8,9weeks between treatment group and control group(P<0.05):treatment group is superior.(3) BMP-2concentration quantitatively:there was statistical significant (P<0.05):treatment group was superior. Conclusion:Application particle gun mediated BMP-2gene to do local gene transfer in therapy of chronic bone defect is efficacious. |
