Objective:To explore changes of intestinal mucosal barrier of rats with severe acute pancreatitis(SAP), and the protection mechanism of Rhubarb.Methods:42male SD rats were randomly divided into three groups,sham operation group(group S, n=10), model group of SAP(group P, n=14) and therapeutic group (group T, n=18).SAP model of rats was induced by injecting adversely5%sodium taurocholate for0.2ml/min(0.1ml/100g) into the common biliary-pancreatic duct.Determined the level of serum amylase,plasma endotoxin and abdominal water after24hours. Observed the pathological changes of pancreas and small intestine through Hematoxylin-eosin staining (HE staining) and the abdominal viscera bacterial translocation rate.Done the quantitative analysis of the intestinal flora use the method of real-time PCR.Results:Compared with group S, in group P, the pancreas and small intestine had severe pathological damage, the pathology score, the plasma endotoxin and the bacterial positive rate were all significantly much higher(P<0.01). Also,the level of E.coli in the cecum contents was much higher (19.43±7.16,P<0.01),while lactobacillus and bifidobacterium were decreased significantly(1.33±0.67,P<0.01or9.17±6.83,P<0.05).Compared with group P,in group T,the pathology score of pancreas and small intestine,the plasma endotoxin and the bacterial positive rate were all significantly lower(P<0.01or P<0.05).And E.coli reduced (15.93±15.21),lactobacilli and bifidobacteria increased (22.88±13.65and11.15±4.66).But there is no statistically significant difference(P>0.05). Conclusion:In SAP rats,the intestinal mucosal barrier was damaged and the intestinal flora was disordered.Rhubarb can relieve the intestinal mucosa injury,shows a certain therapeutic efficacy on SAP.
