| Objective:To research the effect of etomidate emulsion on action potential and ligand-gated chloride ion channel or potassium ion channel of hippocampal CA1. Methods:Decollate40male rats quickly, get out the hippocampus and using the slicing machine to cut out400μm thickness of hippocampus slices.Divided into4groups randomly:lipid emulsion group(n=10);etomidate group(n=10);etomidate+DIDS group(n=10);etomidate+TEA group(n=10).Using current clamp technique to give different stimulus intensity, patch clamp amplifier record the impact of etomidate on action potential firing and ligand-gated chloride ion channel or potassium ion channel. Results: Intralipid did not change the number or amplitude of action potential.As the time incubated by etomidate was postponed,the sum and amplitude of action potential in the soma of the hippocampal CA1neurons obviously reduced.Augmented the stimulus intensity little by little,10μmol/L etomidate added the sum of action potential firing should be notable but the amplitude of action potential had not obviously changed.DIDS+etomidate reduced the sum of action potential after20mins.By the stimulus intensity added,the degenerative time of action potential increased gradually.The last group joined with Tetraethylammonium(TEA) which block potassium ion channel did not affect the suppression function of etomidate.Conclusion:Etomidate emulsion may finally inhibiting acion potential through opening Cl+ion channel in order to increase Cl+internal flow to achieve general anesthesia.K+ion channel may have few effects in this process. |
