Objective:Sleep-disordered breathing seriously endanger people’s health,Presently considered to be the source of a variety of body systemic disease,Being attached great importance to the medical field at home andabroad.Clinically, obstructive sleep apnea syndrome (Obstructive sleepapnea-hypopnea syndrome, OSAHS) the most common. The characteristicof OSAHS is that the patients for a long time experience apnea repeatedlyduring sleep, which leads to repetitive cycles of hypoxia and reoxygenation,which led to patients with multiple systems and/or organ damage.to studythe effectsof chronic Intermittent hypoxia-induced inflammatory cytokinesand removal of hypoxia on on GLUT-4of skeletal muscle expression inSprague-Dawley rats, and to furnish the theoretic foundation andexperimental evidence for the treatment of patients with OSAHS,who hasbeen associated with disorders of glucose metabolism.Methods:A total of24Sprague-Dawley male rats were randomly assigned tothree experimental groups (n=8/group): unhandled control(UC) group, chronic intermittent hypoxia (CIH) group, and removal of hypoxia(RH)group. At the end of the experiment,fasting blood glucose(FBG),fastingblood insulin (FINS)and serum inflammatory cytokines levels wereseparatelymeasured by glucose oxidase-peroxidase、 InsulinRadioimmunoassay Kit and ELISA kits. Homeostasis model assessment(IRI) was used to measure insulin resistance in each animal. Westernblotting were used to measure GLUT-4protein in skeletal muscle tissue.Results:①When compared with UC group, CIH increased fasting bloodglucose、 blood insulin levels and IRI (P<0.05), fasting blood glucose inRH group was higher than those in UC group (P<0.05).②inflammatory cytokines levels(IL-6、TNF-α) in CIH group washigher than those in UC and RH group (P<0.05), RH group was higher thanUC group.③The GLUT-4protein expression in skeletal muscle tissue wassignificantly reduced after CIH compared with UC (P<0.05),and wasimproved after RH compared with CIH group (P<0.05).Conclusions:CIH in adult rats could induce inflammatory cytokines increases andinsulin resistance, insulin resistance is associated with decreased GLUT-4protein expression of skeletal muscle tissue,which induced by inflammatorycytokines.
